摘要
目的:研究黄柏水溶液对大鼠下肢静脉性溃疡创面修复中NF-κB、TNFAIP3/A20表达的影响。方法:将雄性SD大鼠80只随机平均分为假手术组、模型组、治疗组(黄柏水溶液)和对照组(复方黄柏液)。每组又分为7d和14d两个时间点,每个时间点10只。分别于治疗第7天,第14天采用数码相机照相电子计算机软件分析及透明生物膜描膜法观察溃疡创面愈合情况。治疗第7、14天进行血流变及DD检测,ELISA法检测血清中IL-1β、IL-6和TNF-α水平。取创面组织进行病理学检测,同时用PCR检测NFκB、IκB、TNFAIP3的表达。结果:治疗组与模型组比较,效果明显,愈合率显著升高,溃疡面积不断缩小,差异有统计学意义(P<0.05);与模型组比较,IL-1β、IL-6、TNF-α的含量降低明显,差异有统计学意义(P<0.05);与模型组比较NF-κB mRNA水平下降,IκBα、TNFAIP3/A20的水平上调,差异有统计学意义(P<0.05)。治疗14天的效果要优于药物治疗第7天。各项指标结果显示,对照组于治疗组没有明显差异。结论:黄柏水溶液抑制了大鼠下肢静脉性溃疡创面修复中的NF-κB的表达,促进了TNFAIP3/A20的表达。
Objective:To investigate the wound-healing effect of aqueous solution of Cortex Phellodendri on the expressions of NF-κβand TNFAIP3/A20in rats with venous ulcer of lower limbs.Methods:80male SD rats were randomly divided into the sham surgery group,the model group,the observation group(aqueous solution of Cortex Phellodendri),and the control group(Compound Phellodendron Liquid).Each group was subdivided by7d and14d,10rats at each time point.Wound-healing degree was observed,tests of hemorheology DD were performed,serum levels of IL-1β,IL-6,and TNF-a,as well as the expressions of NF-κβand TNFAIP3/A20were detected on7th and14th day respectively.Results:The effect of the observation group was more obvious,the healing rate increased more significantly,the area of the ulcer was narrower,compared to the model group,and the difference was statistically significant(P<005).The levels of IL-1β,IL-6,and TNF-αwere decreased significantly,and the difference was statistically significant,compared to the model group(P<005).The expression of NF-κβwas reduced,but TNFAIP3/A20was raised when compared to the control group,with a significant difference(P<0.05).14d treatment was better than7d treatment.It showed that there were no obvious difference between the observation group and the control group in the above indicators.Conclusion:Aqueous solution of Cortex Phellodendri can inhibit the NF-κβexpression and promote TNFAIP3/A20expression,which may be related to its wound-healing effect in rats with venous ulcer of lower limbs.
