摘要
目的:探讨miR-204在肺癌组织中的表达与临床病理的关系。方法:选择行肺癌切除术的78例石蜡存档组织标本进行研究,取癌症组织作为试验组,取癌旁正常组织作为对照组。采用荧光定量实时PCR(RT-PCR)对肺组织miR-204荧光相对表达量进行检测,并分析其荧光相对表达量和性别、年龄、肿瘤大小、淋巴结转移、TNM分期、分化程度、组织类型的关系。结果:试验组miR-204表达荧光相对含量水平较对照组显著降低(P<0.05);miR-204表达荧光相对含量在性别、年龄方面比较,差异无统计学意义(P>0.05),而在病程、肿瘤长径、淋巴结转移、TNM分期、分化程度和组织类型等不同临床病理条件下其荧光相对含量水平差异有统计学意义(P<0.05)。结论:肺癌组织中miR-204表达荧光相对免疫含量较正常组织明显降低,且病程越长、肿瘤越大,淋巴结转移、TNM≥Ⅲ期、低分化和非小细胞肺癌组织中其表达水平均较低,推测miR-204靶基因对肺癌发生、恶化和进展存在显著调控作用。
Objective:To discuss the expression of miR-204in the lung cancer tissues and the relationship between it with the clinical pathology.Methods:Seventy-eight cases of paraffin archived tissue specimens of patients who were treated with resection of lung cancer in our hospital were selected to conduct the research,and the lung cancer tissues were selected as the experimental group.while the tumor adjacent normal tissues were selected as the control group.The fluorescence quantitative real-time PCR(RT-PCR)was used to detect the relative expressions of miR-204in lung tissues,and the relationships between relative expressions with the gender,age,tumor size,lymph node metastasis,TNM stage,degree of differentiation,tissue types were analyzed.Results:The relative content of miR-204expression in the experimental group was significantly lower than that in the control group(P<0.05).There was no significant difference in the relative content of miR-204expression between sex and age(P>0.05),the differences of fluorescence relative content between different clinical pathological conditions of course of disease,length of tumor,lymph node metastasis,TNM stage,degree of differentiation and histological type were statistically significant(P<0.05).Conclusion:The relative contents of miR-204expression in the lung cancer tissues were obviously lower than those in the normal tissues,and in patients with long duration,large tumor bigger,lymph node metastasis and TNM stageⅢand above,and poorly differentiated non-small cell lung cancer,the expression levels were low,so it was suggested that the miR-204target genes had significant regulation and control roles on the development and progression of lung cancer.
