摘要
目的研究前列通片对前列腺癌的体内和体外抑制效果。方法选用3株前列腺癌肿瘤细胞对前列通片含药血清进行体外抑瘤试验。根据体外试验筛选PC-3细胞株,制作80只的PC-3荷瘤裸鼠。筛选肿瘤直径约100 mm,相对均一的50只荷瘤裸鼠,随机分为模型(溶媒)对照组、阳性对照组、前列通低、中、高剂量组,10只/组。每只动物按照20 m L/kg体积灌胃给药,1次/d,连续32 d。结果体外筛选:前列通片含药血清(浓度为20%,10%,5%)对PC-3细胞存活率分别为47.6%、87.2%、112.9%;对22 RV1细胞的存活率分别为90.9%、89.6%、85.3%,对DU145细胞的存活率分别为82.7%、106.3%、100.4%。体内实验:与模型对照组比较,前列通片低剂量组肿瘤重量未见显著降低,肿瘤生长抑制率18.5%,前列通片中、高剂量组及阳性对照组肿瘤重量显著降低(P<0.05),肿瘤生长抑制率分别为40.3%、23.8%、62.6%。结论在本实验条件下,前列通片含药血清对PC-3细胞抑制效果较为明显。2.04 g/kg的前列通片对PC-3前列腺癌裸鼠移植瘤有一定的抑制作用。
Objective To observe the effect of Qianlietong tablets on inhibition of tumor cell growth.Methods Observe the inhibitive effect in vitro of the3strains of prostate cancer cells with Qianlietong tablets containing serum.According to the experimental results,80PC-3tumor bearing nude mice were produced.50tumor bearing nude mice which the tumor diameter was about100mm were randomly divided into model control group,positive control group,Qianlietong low,medium and high dose groups,10rats in each group.Each animal was administered orally in accordance with the volume of20mL/kg,1times/day,32days in a row.Results Qianlietong tablets containing serum(concentration of20%,10%,5%)on PC-3cell survival rate were47.6%,87.2%,112.9%,on22RV1cell survival rate were90.9%,89.6%,85.3%,on DU145cell survival rate were82.7%,106.3%,100.4%.Compared with the model control group,the tumor growth inhibition rate of the low dose group was18.5%.The tumor weight of the high dose group and the positive control group was significantly decreased(P<0.05).The tumor growth inhibition rate was40.3%,23.8%,62.6%,respectively.Conclusion Under the experimental conditions,Qianlietong tablets containing serum on PC-3cell inhibitory effect is more obvious.Qianlietong tablets has a certain inhibitory effect on the prostate cancer with PC-3in nude mice.
