摘要
为开发高效安全的对虾黑变抑制剂,研究3种巯基化合物对凡纳滨对虾(Litopenaeus vannamei)多酚氧化酶(Polyphenol oxidase,PPO)的抑制效果和动力学。结果表明:当L-半胱氨酸、N-乙酰-L-半胱氨酸和还原性谷胱甘肽质量浓度分别为0.4、0.8、1.6 mg/mL时,与PPO作用5 min,对PPO的抑制率分别达85.3%、89.3%、91.2%;3种巯基化合物质量浓度为0~0.8 mg/mL时对凡纳滨对虾PPO的抑制为竞争性抑制,质量浓度为0.8~6.4mg/mL时的抑制为不可逆抑制。3种巯基化合物对凡纳滨对虾PPO的抑制是两种机制并存。
In order to develop safe and effective inhibitors of shrimp melanosis,inhibition effects and kinetics of three sulfhydryl compounds on polyphenol oxidase from Litopenaeus vannamei were studied.When the concentrations of L-cysteine,N-acetyl-L-cysteine and reduced glutathione were0.4,0.8and1.6mg/mL and reacted with PPO for5min,the inhibition rates were85.3%,89.3%and91.2%,respectively.Electrophoresis and inhibition kinetics revealed that the sulfhydryl compounds showed a competitive inhibition at the concentrations of0-0.8mg/mL and showed an irreversible inhibition at the concentrations of0.8-6.4mg/mL.Therefore,the two inhibitory mechanisms of three sulfhydryl compounds on PPO may be coexistent.
