急性心肌梗死大鼠主动脉组织中Profilin-1蛋白表达与细胞损伤、凋亡及ERK1/2信号通路关联研究 被引量:8

Expression of Profilin-1 protein in aortic tissue and correlation among it and cell injury, apoptosis and ERK1/2 signaling pathway in rats with acute myocardial infarction

摘要 目的探讨Profilin-1蛋白在急性心肌梗死大鼠主动脉组织中的表达及其与细胞损伤、凋亡及ERK1/2信号通路的关系。方法 20只成年雄性Wistar大鼠随机分为心肌梗死模型组和假手术对照组,每组各10只。对两组大鼠进行急性心肌梗死模型处理。Western免疫印迹法测定两组大鼠急性心肌梗死后升主动脉组织内Profilin-1蛋白和细胞外信号激酶磷酸化蛋白(p ERK1/2)的表达量,流式细胞术检测心肌细胞内皮微颗粒(EMP),分光光度计法测定一氧化氮(NO)水平、心肌损伤指标肌钙蛋白T(c Tn T)和肌酸激酶同工酶(CK-MB)水平,RT-PCR法检测心肌凋亡因子P53、Fas、Bax、Bcl-2 m RNA的表达水平。结果与对照组相比,模型组大鼠的主动脉组织Profilin-1和p ERK1/2蛋白表达水平较高(P<0.05);模型组EMP值较高,NO水平较低(P均<0.05);心肌组织中c Tn T、CK-MB、P53、Fas和Bax因子m RNA表达较高,Bcl-2 m RNA水平较降(P均<0.05)。模型组Profilin-1和p ERK1/2蛋白表达量与c Tn T、CK-EMP、P53、Fas和Bax水平呈正相关,与NO和Bcl-2水平呈负相关(P均<0.05)。结论在大鼠急性心肌梗死模型中Profilin-1蛋白表达升高。 Objective To investigate the expression of Profilin-1protein in aortic tissue and correlationamong it and cell injury,apoptosis and ERK1/2signaling pathway in rats with acute myocardial infarction(AMI).Methods Adult male Wistar rats(n=20)were randomly divided into model group and control group(each n=10),and AMI model was established.The expressions of Profilin-1protein in aortic tissue and extracellular pERK1/2protein were detected by using Western blotting,and myocardial endothelial microparticles(EMP)were detectedby using flow cytometry.The levels of nitric oxide(NO),cardiac troponin T(cTnT)and creatine kinase MB(CKMB)were detected by using fluorospectrophotometry.The expressions of P53mRNA,Fas mRNA,Bax mRNAand Bcl-2mRNA were detected by using reverse transcription-polymerase chain reaction(RT-PCR).ResultsThe expressions of Profilin-1and pERK1/2were higher in AMI group than those in control group(P<0.05).Inmodel group,EMP was higher and NO was lower(all P<0.05).The expressions of cTnT,CK-MB,P53mRNA,Fas mRNA and Bax mRNA were higher and expression of Bcl-2mRNA was lower in model group(all P<0.05).The expressions of Profilin-1and pERK1/2were positively correlated to cTnT,CK-EMP,P53,Fas and Bax,andnegatively correlated to NO and Bcl-2in model group(all P<0.05).Conclusion The expression of Profilin-1protein increases in rat model of AMI.
