‘三红蜜柚’果实类胡萝卜素合成途径部分酶基因的克隆及表达分析 被引量:6

Cloning and Analysis of Partial Enzyme Genes of Carotenoid Biosynthetic Pathway from Citrus maxima cv. 'Sanhongmiyou'

摘要 果实颜色是柚(Citrusmaxima)的重要性状之一,为了解其色泽调控机理,本研究以‘三红蜜柚’为材料,从柚转录组数据库(未发表)中筛选得到CmPSY、CmPDS、CmZDS和CmLCYB的序列,分别编码438、533、570、214个氨基酸。生物信息学表明,CmPSY、CmPDS、CmZDS和CmLCYB编码的氨基酸序列与可可树、木薯和甜橙的同源相似度较高,尤其是与甜橙的同源性分别高达97.95%、99.81%、97.89%和100%。实时荧光定量PCR检测结果表明,CmPSY、CmPDS、CmZDS和CmLCYB在蜜柚的汁胞、囊衣和海绵层中均有表达,且表达量基本都呈先增后降的趋势,说明这些基因在蜜柚类胡萝卜素代谢中不可或缺。4个基因表达水平基本都在花后150~180 d达到最大,说明这些基因之间有着密切的协作关系。本研究为类胡萝卜素在蜜柚果实各个组织的调控提供了研究信息,并为进一步通过基因调控来提高蜜柚的外观品质奠定了基础。 Fruit color is an important trait for Citrus maxima. In order to insight into the molecular basis of color, C. maxima cv. ‘Sanhongmiyou' was used as the experimental material in this study, and the sequences of CmPSY, CmPDS, CmZDS and CmLCYB were chosen from the data of transcriptome(unpublished), which respectively encoded 438, 533, 570 and 214 amino acids. Bioinformatics analysis showed that the amino acids of CmPSY, CmPDS, CmZDS and CmLCYB were highly homologous to the same amino acids of Theobroma cacao, Manihot esculenta and Citrus sinensis, and the closest homolog is C. sinensis, with 97.95%, 99.81%, 97.89% and 100% amino acid identity. Quantitative real-time PCR analysis showed that CmPSY, CmPDS, CmZDS and CmLCYB were expressed in the juice sacs, segment membrane and albedo of C. maxima. And the amount of expression basically increased first and then decreased, which indicating that the genes of these enzyme were indispensable in the metabolism of carotenoid in C. maxima. And the expression levels of the four genes reached maximum at 150 to 180 days after flowering, which indicating that there was a close collaboration between them. This study would provide the research information for the regulation of carotenoids in various organizes of C. maxima, and it also lay the foundation for further gene regulation to improve the appearance quality of C. maxima.
