摘要
目的检测口腔鳞状细胞癌(oral squamous cell carcinoma,OSCC)患者外周血中白细胞介素32(interleukin-32,IL-32)的表达水平,探讨其在OSCC发病中的临床意义。方法收集55例OSCC患者(口腔癌组)及35例健康志愿者(对照组)的外周血进行检测,外周血细胞总RNA用Trizol试剂,然后将总RNA反转录为c DNA,再用real-time PCR检测IL-32 mRNA的表达水平;外周血IL-32蛋白质表达水平用酶联免疫吸附试验(enzyme-linked immunosorbent assay,ELISA)检测; Pearson相关性分析IL-32表达水平与肿瘤发生、发展、转移的相关性。结果 OSCC患者外周血中IL-32 mRNA及蛋白质表达均显著高于对照组(P <0. 05)。Pearson相关性分析显示,OSCC患者外周血中IL-32表达水平分别与OSCC临床分期、淋巴结转移呈正相关(P <0. 05)。结论 OSCC患者外周血中IL-32呈明显的高表达,其可能参与OSCC的发病,是判断OSCC发生发展的重要指标之一。
Objective To investigate the level of serum interleukin-32（IL-32） and its significance in patients with oral squamous cell carcinoma（OSCC）. Methods Fifty-five patients with oral squamous cell carcinoma were enrolled in the present study. Thirty-five healthy individuals were enrolled as the control group. Peripheral blood of patients and healthy individuals was collected,respectively. Total RNA of PBMCs was extracted using Trizol. Then RNA was reversely transcribed into c DNA using reverse transcription Kit. Subsequently,IL-32 mRNA expression was detected using real-time PCR. Serum IL-32 protein expression was measured by enzyme-linked immunosorbent assay（ELISA）. Then the relations of IL-32 to tumorigenesis,development and metastasis were analyzed by using Pearson＇ s correlation analysis method, respectively.Results The levels of IL-32 mRNA and protein in peripheral blood of OSCC patients were higher than that in the control group（P〈0. 05）. Pearson＇s correlation analysis showed that serum expression of IL-32 in patients with OSCC was positively correlated with tumorigenesis,development and metastasis of OSCC（P〈0. 05）,respectively. Conclusion Serum IL-32 expression rises in patients with OSCC. So we believe that IL-32 might be involved in the pathogenesis of OSCC.
