同源四倍体及原二倍体黑皮冬瓜抗逆相关指标及同工酶比较 被引量:5

Comparison on Indices Related to Stress Resistance and Isoenzyme of Autotetraploid and Diploid Benincasa hispida (Thunb.) Cogn.

摘要 以同源四倍体及其原二倍体黑皮冬瓜为试验材料,对其抗逆性相关指标及同工酶进行了比较研究。结果表明:同源四倍体黑皮冬瓜叶片中超氧化物歧化酶(SOD)活性在抽蔓期和开花期极显著高于原二倍体(P<0.01),果实膨大期略高于原二倍体(P>0.05);过氧化氢酶(CAT)活性在抽蔓期和果实膨大期略高于原二倍体(P>0.05),开花期略低于原二倍体(P>0.05);过氧化物酶(POD)活性在抽蔓期和果实膨大期略高于原二倍体(P>0.05),开花期则显著高于原二倍体(P<0.05);丙二醛(MDA)含量在抽蔓期略低于原二倍体(P>0.05),开花期和果实膨大期极显著低于原二倍体(P<0.01);游离脯氨酸(Pro)含量在3个生育期内均略高于原二倍体(P>0.05);可溶性蛋白质含量在抽蔓期极显著高于原二倍体(P<0.01),在开花期和果实膨大期略高于原二倍体(P>0.05)。在开花期同源四倍体黑皮冬瓜叶片过氧化物酶(POD)同工酶和酯酶(EST)同工酶均比原二倍体多1条酶带,超氧化物歧化酶(SOD)同工酶比原二倍体少2条酶带,且酶带的宽度和着色程度较原二倍体深。同源四倍体黑皮冬瓜植株潜在的抗逆能力优于原二倍体,同源四倍体黑皮冬瓜植株酶的种类与原二倍体相似,但剂量表达量与原二倍体存在明显差异。 Taking autotetraploid and diploid Benincasa hispida (Thunb.)Cogn.as materials,the indices related to stress resistance and isoenzyme were studied.The results showed that the activity of SOD of autotetraploid Benincasa hispida (Thunb.)Cogn.was significantly higer than diploid in vine growth stage and in flower growth stage (P〈0.01),and slightly higher than diploid in fruit growth stage (P〉0.05);the activity of CAT of autotetraploid Benincasa hispida (Thunb.)Cogn.was slightly higher than diploid in vine growth stage and in fruit growth stage (P〉0.05),and slightly lower than diploid in flower growth stage (P〉0.05);the activity of POD of autotetraploid Benincasa hispida (Thunb.)Cogn.was slightly higher than diploid in vine growth stage and in fruit growth stage (P〉0.05),and significantly higher than diploid in flower growth stage (P〈0.05);the MDA content of autotetraploid Benincasa hispida (Thunb.)Cogn.was slightly lower than diploid in vine growth stage (P〉0.05),and significantly lower than diploid in flower growth stage and in fruit growth stage (P〈0.01);the Pro content of autotetraploid Benincasa hispida (Thunb.)Cogn.was slightly higher than diploid in three different growth stage (P〉0.05);the soluble protein content of autotetraploid Benincasa hispida (Thunb.)Cogn.was significantly higher than diploid in vine growth stage (P〈0.01),and slightly higher than diploid in flower growth stage and in fruit growth stage (P〉0.05).The POD and EST isoenzyme bands of autotetraploid were all one more than those of diploid in leaves in flower growth stage,and the SOD isoenzyme bands of autotetraploid was two less than diploid,but the expressed quantity of autotetraploid was higher than diploid.The results indicated the autotetraploid Benincasa hispida (Thunb.)Cogn.presented great potential in stress resistance,and the isoenzyme band pattern of autotetraploid and diploid were the same,but the activities in autotetraploid were stronger than that in diploid in leaves.
