摘要
目的建立地龙及酒制地龙氨基酸类指纹图谱,并测定5种氨基酸的含有量。方法分析采用Diamonsil C_(18)色谱柱(250 mm×4. 6 mm,5μm);以乙酸钠缓冲液-乙腈为流动相,梯度洗脱;体积流量0. 8 mL/min;检测波长360 nm;柱温27℃。结果 18批生地龙、酒地龙样品均有19个共有峰,平均相似度分别为0. 921、0. 991。甘氨酸、丙氨酸、缬氨酸、亮氨酸、赖氨酸分别在1. 34~840、0. 79~491、0. 87~544、0. 74~461、0. 51~315μg/mL范围内线性关系良好(r≥0. 999 5),平均加样回收率(RSD)分别为95. 1%(1. 87%)、94. 0%(1. 86%)、92. 0%(0. 98%)、88. 7%(3. 56%)、96. 8%(1. 23%)。结论该方法稳定可靠,可用于地龙的质量控制。
AIM To establish the amino acid fingerprints of Pheretima and Pheretima processed by wine, and to determine the contents of five kinds of amino acids constituents. METHODS The analysis was devel- oped on a 27℃ thermostatic Diamonsil C18 (250 mm × 4.6 mm, 5 μm) , with the mobile phase comprising of sodium acetate buffer-acetonitrile flowing at 0.8 mL/min in a gradient elution manner, and the detection wave- length was set at 360 nm. RESULTS There were nineteen common peaks in the fingerprints of eighteen bat-ches of samples for Pheretima, Pheretima processed by wine, with the similarities of 0. 921 , 0. 991, respectively. Glycine, alanine, valine, leucine and lysine showed good linear relationships within the ranges of 1.34 - 840, 0.79-491, 0.87 -544, 0.74 -461, 0.51 -315 μg/mL (r≥0.999 5), whose average recoveries (RSDS) were95.1% (1.87%), 94.0% (1.86%), 92.0% (0.98%), 88.7% (3.56%), 96.8% (1.23%) , respectively. CONCLUSION This stable and reliable method can be used for the quality control of Pheretima.
