摘要
目的建立采用超声提取,高效液相色谱法(HPLC)测定山葡萄藤中白藜芦醇的含量。方法采用色谱柱:Inertsil ODS-3 C18色谱柱(250 mm×4.6 mm,5μm);流动相:甲醇-0.05%磷酸溶液(40:60);流速:1.0 mg/m L;柱温:30℃;检测波长:306 nm;进样量:10μL。结果白藜芦醇在0.504~75.6 mg/m L范围内呈良好的线性关系。测定10批山葡萄藤中白藜芦醇平均含量为0.48%,平均回收率95.16%(RSD=1.94%,n=9)。结论该实验采用超声提取,并对提取条件进行探讨,得出最佳条件为超声时间45 min、超声功率300 W、固液比1∶250(g/m L)、溶剂为甲醇。该研究表明该方法简便、快速、经济、可靠,专属性强,可用于山葡萄藤的质量控制。
Objective Ultrasonic extraction and high performance liquid chromatography(HPLC) were used to determine the content of resveratrol in grape vines. Methods Chromatography column: inertsil ODS-3 C18 column(250 mm×4.6 mm,5 μm);mobile phase: methanol-0.05% phosphoric acid solution(40:60); flow rate: 1.0 m L/min; column temperature : 30°C; detection wavelength: 306 nm; injection volume: 10 μL. Results Resveratrol showed a good linear relationship in the range of0.504 to 75.6 mg/m L. The average content of resveratrol in 10 batches of vines was 0.48%, and the average recovery was95.16%(RSD=1.94%,n=9). Conclusion Ultrasonic extraction was used in this experiment and the extraction conditions were investigated. The optimal conditions were: ultrasonic time 45 min, ultrasonic power 300 W, solid-liquid ratio 1:250(g/m L),and methanol as the solvent. The study shows that this method is simple, rapid, economical, reliable, and highly specific. It can be used for quality control of vine vines.
