两种大鼠肝纤维化模型的病理特点及影像学研究 被引量:3

A comparative study suitable for imaging studies of liver fibrosis in rats

摘要 目的:比较适用于影像学研究的四氯化碳法和胆管结扎法所致两种大鼠肝纤维化模型的肝脏病理学变化。方法:四氯化碳法为皮下注射50%的四氯化碳橄榄油溶液0.3 m L·(100 g)-1,每周2次;胆管结扎法为麻醉后手术,在胆管近端和远端2处结扎胆管。于制模后不同时间点取出大鼠行磁共振扩散加权成像及病理等检查。结果:获得各个时期的大鼠肝纤维模型(F0=16只;F1=16只;F2=16只;F3=17只;F4=15只)。磁共振ADC值在纤维化各期不同程度减低,且在各病理分期存在差异。四氯化碳组肝脏气球样变及脂肪变性于F2期达到顶峰。肝实质炎症改变F4期炎症最为明显。而胆管结扎组肝脏气球样变及脂肪变性不甚明显。但炎症评分高于四氯化碳组。两组的胶原面积均随着纤维化分期进展而增高。同一分期的模型中,胆管结扎组的胶原面积高于四氯化碳组。结论:两种方法都可诱发大鼠肝脏发生纤维化,诱发不同的时间,可获得肝纤维不同病理阶段的动物模型,且适合磁共振研究。可根据不同的实验目的选择不同的方法建模。 Objectives: To compare pathological changes in the liver fibrosis caused by carbon tetrachloride(CCl4) and bile duct ligation(BDL). Methods: The rats in the CCl4 group received 50% CCl4(CCl4: olive oil,1∶ 1) subcutaneously at a dose of 0. 3 m L·(100 g)-1 of body weight,twice a week. In the BDL group common bile duct was isolated and double-ligated. At the different time points after the model was taken out,magnetic resonance imaging and pathological examinations were performed. Results: Different stages of liver fibrosis were obtained(F0 = 16; F1 = 16; F2 = 16;F3 = 17; F4 = 15). Mean ADC values decreased with increasing fibrosis levels. In CCl4 group,the highest percentage of steatosis and greatest hepatocyte ballooning were observed in stage F2 specimens. High levels of inflammation were observed in livers with stage F4 fibrosis. In the BDL group,hepatic ballooning and steatosis were not obvious. The inflammation score was higher than that of the CCl4 group. In the same stage of the model,the collagen area of the BDL group was higher than that of the CCl4 group. Conclusions: Both methods can induce liver fibrosis in rats,induce different time,obtain different stages of liver fibrosis,and suitable for magnetic resonance studies. Different methods can be selected for modeling according to different purposes.
