褐飞虱卵巢特异性基因Nl10847的功能初探 被引量:3

摘要 褐飞虱Nilaparvata lugens(Stl)是水稻最主要的害虫,其繁殖力高低是影响水稻产量损失最直接的因素。本文旨在研究褐飞虱卵巢特异性基因Nl10847对褐飞虱繁殖力的影响。首先通过克隆验证了Nl10847的开放阅读框(ORF)为552 bp,编码184个氨基酸;然后利用q-PCR技术分析其时空表达模式,发现其在卵巢组织和成虫期高表达;最后通过RNAi的方法,发现下调Nl10847基因的表达后会抑制全虫和卵巢中NlVgR的mRNA表达水平,并严重阻碍卵巢的发育。这些结果表明Nl10847在卵巢发育成熟过程中承担重要的功能。 The brown planthopper(BPH) Nilaparvata lugens(Stl) is one of the most serious insect pests of rice. Its fecundity is the direct factor determining the extent of damage to rice. This paper aims to study the effects of ovarian specific gene Nl10847 on the fecundity of BPH. Firstly,the open reading frame(ORF) of Nl10847 was cloned and verified to be 552 bp, encoding 184 amino acids.Subsequently,q-PCR was used to analyze the spatiotemporal expression patterns,and high expression was found in ovarian tissue and adult stage. Finally,by using RNAi,it was found that the downregulated expression of Nl10847 inhibited the mRNA expression level of NlVgR in the whole insect and the ovary,and seriously delay ovary development of the insects. These results suggest that Nl10847 play an important role in ovarian development and maturation.
