摘要
该研究以实验室构建的重组大肠杆菌E.coli BL21/p ET28a(+)-mle为研究对象,以L-乳酸量为评价指标,通过单因素和正交试验研究了诱导温度、培养基起始pH值、异丙基-β-D-硫代吡喃半乳糖苷(IPTG)浓度和诱导时间对重组大肠杆菌E.coli BL21/p ET28a(+)-mle分泌表达苹果酸乳酸酶(MLE)的影响。结果表明,重组大肠杆菌E.coli BL21/p ET28a(+)-mle分泌表达苹乳酶的最佳培养条件为培养基起始pH值为5.9,诱导温度为28℃,IPTG浓度0.6 mmol/L,诱导时间3 h。在此优化条件下,L-乳酸产量为2.37 g/L,较优化前提高9.48倍。
Using the constructed recombinant E. coli BL21/p ET28 a(+)-mle as research object, and L-lactic acid production as evaluation index, the effects of inducing temperature, medium initial pH, isopropyl-β-D-1-thiogalactopyranoside(IPTG) concentration, and inducing time on malolactic enzyme production secreted and expressed by E.coli BL21/p ET28 a(+)-mle were studied using single factor and orthogonal experiments. The result showed that the optimal culture conditions for the secretory and expression of malolactic enzyme by recombinant E. coli BL21/p ET28 a(+)-mle were:initial medium pH 5.9, inducing temperature 28 ℃, IPTG concentration 0.6 mmol/L, and inducing time 3 h. Under the optimized condition, the yield of L-lactic acid was 2.37 g/L, which was 9.48 times higher than that of before optimization.
