摘要
目的研究micro RNA-32(miR-32)在肾透明细胞癌(SE)患者肿瘤组织中表达及与SE病理分级的关系。方法选择76例于青海大学附属医院泌尿外科收治的SE患者,其中男性48例,女性28例;年龄38~65岁,平均年龄53.44岁;TNM分期,Ⅰ期14例,Ⅱ期20例,Ⅲ~Ⅳ期42例;肿瘤直径<4 cm 44例,≥4 cm 32例。取手术后病灶组织和癌旁组织,采用实时荧光定量聚合酶链反应检测miR-32、miR-21在肿瘤组织及癌旁组织中的表达,分析miR-32与SE患者病理特征的关系;采用Pearson相关性检验分析miR-32与miR-21相关性;采用Western blot检测SE组织及癌旁组织中纤维状肌动蛋白(F-actin)、磷酸酶张力蛋白同源基因(PTEN)、波形蛋白(vimentin)的表达。结果 SE患者肿瘤组织中miR-32、miR-21表达明显高于癌旁组织(P<0.05);与肿瘤直径<4 cm相比,SE患者肿瘤直径≥4 cm肿瘤组织中miR-32表达升高,且随TNM分期升高而升高。Pearson相关性分析结果显示肿瘤组织中miR-32与miR-21表达呈正相关(r=0.627,P<0.05)。与癌旁组织相比,SE肿瘤组织中F-actin、vimentin表达升高,PTEN表达降低(P<0.05)。结论 miR-32在SE肿瘤组织中呈高表达,并且与肿瘤直径、临床分析密切相关,其机制可能与miR-32可促进SE细胞迁移有关。
Objective To study the expression of micro RNA-32( mi R-32) in tumor tissues of patients with suprarenal epithelioma(SE) and the relationship with pathological grade of SE. Methods A total of 76 SE patients undergoing urology treatment were enrolled, which included 48 males and 28 females, aged 38-65 years old with mean age of 53.44 years old. In TNM staging, 14 cases of stage Ⅰ, 20 of stage Ⅱ and 42 of stage Ⅲ-Ⅳ; 44 cases of tumor with diameter 4 cm and 32 cases 4 cm. Real-time PCR was used to detect the expression of mi R-32 and mi R-21 in post-operative lesion tissue and paracancerous tissue. The relationship between mi R-32 and pathological features of SE was analyzed. Pearson correlation test was used to analyze the correlation between mi R-32 and mi R-21; Western blot was used to detect the expression of fibrillar actin(F-actin), phosphatase tension protein homologue(PTEN) and vimentin(vimentin) in SE tissues and adjacent tissues.Results The expression of mi R-32 and mi R-21 in tumor tissues of the SE patients was significantly higher than that in ad-jacent tissue(P 0.05). Compared with tumor diameter 4 cm, the expression of mi R-32 in tumors with diameter ≥ 4 cm increased in SE patients, with the increase of the TNM staging; Pearson correlation analysis showed that there was a positive cor-relation between the expression of mi R-32 and mi R-21 in tumor tissues(r = 0.627, P 〈0.05). Compared with the adjacent tissue, the expression of F-actin and vimentin was increased and the expression of PTEN was decreased in SE tumor tissues( P 〈0.05). Conclusion It is demonstrated that mi R-32 is highly expressed in tumor tissue of SE and is closely related to tumor diameter and clinical analysis. The possible mechanism may be that mi R-32 promotes the migration of SE cells.
