中国罗非鱼主养区无乳链球菌流行菌株的菌毛岛屿及其血清型分型 被引量:8

The pilus islands and molecular serotype of Streptococcus agalactiae isolated from tilapia in the major tilapia-culturing areas in China

摘要 为了解近年来中国罗非鱼主养区无乳链球菌流行特征,本研究通过PCR扩增和测序等方法对2007年~2016年分离纯化得到的263株罗非鱼无乳链球菌进行菌毛岛屿和血清型分型分析。菌毛岛屿分型结果显示:263株罗非鱼无乳链球菌可分为两种菌毛岛屿类型:PI-2b型(40株)和PI-1+PI-2b型(218株),其中PI-1+PI-2b型无乳链球菌是主要的流行菌株,占总数的82.9%。其余5株无乳链球菌的菌毛岛屿基因缺失,不含菌毛岛屿。分子血清型分析结果显示,所有的无乳链球菌分为两种血清型:Ia型258株(98.1%)和Ib型5株(1.9%)。所有Ib型无乳链球菌的菌毛岛屿均有缺失,仅含有2b-AP1基因。进一步分析表明,Ia-(PI-2b)型无乳链球菌是2011年之前主要的流行株型,而Ia-(PI-1+PI-2b)型无乳链球菌为近6年来广泛流行的株型。Ia型是我国罗非鱼无乳链球菌最流行的分子血清型,同时PI-2b是最重要的菌毛岛屿(98.1%)。本研究为罗非鱼无乳链球菌的流行病学和疾病防控提供调查依据。 The aim of this study is to analyze the molecular characteristics of Streptococcus agalactiae isolated from tilapia in the major tilapia-eulturing areas in China. In the present study, a total of 263 S.agalactiae isolates were subjected to examination of molecular serotype and pilus islands by PCR and sequencing. The results showed that the 218 S.agalactiae possessed the PI-I+PI-2b pilus islands and 40 S.agalactiae had the PI-2b pilus islands, and the remaining 5 S.agalactiae had no pilus islands. Molecular serotype detection indicated that the 258 S.agalactiae (98.1%) were serotype Ia, and the remaining 5 S.agalactia (1.9%) were lb. In addition, all lb S.agalactiae were negative for pilus island detection, which only carried 2b-AP1 gene. Importantly, Ia- (PI-1) S.agalactiae was the predominant before 2011, but since then the Ia-(PI-l+PI-2b) S.agalactiae was the predominant instead. In conclusion, serotype Ia S.agalactiae was the most prevalence serotype in China, and the most important pilus islands were the PI-2b (98.1%). This study provided a basis framework for epidemiological analysis and prevention of the streptococcosis in tilapia.
