2009─2017年福建省猪繁殖与呼吸综合征病毒分子流行病学调查和遗传变异分析 被引量:17

Genetic diversity and evolutionary characteristics of PRRSV in Fujian Province from 2009 to 2017

摘要 为了了解福建省猪繁殖与呼吸综合征病毒(PRRSV)流行动态和遗传变异特性,应用RT-PCR方法对福建省龙岩市、漳州市、厦门市、福州市、三明市、莆田市和宁德市等七市的102个不同规模的猪场开展了PRRSV病原学调查。基于ORF5基因的系统进化分析表明,福建省Type 2 PRRSV可分为lineage 1、lineage 3、lineage 5.1和lineage 8.7四个谱系,其中lineage 1(NADC30-like PRRSV)和lineage 3是福建省新出现的毒株。2009—2013年福建省以lineage 8.7的毒株为主,2014—2017年流行毒株发生了转变,以NADC30-like PRRSV为主。PRRSV GP5中和表位发生较高程度的变异,N-糖基化位点增多并逐步靠近下游的中和表位。结果表明,福建省PRRSV呈现复杂多样性,猪场防控PRRS应该采取综合防控措施,避免引入新毒株。 In order to assess the genetic diversity of porcine reproductive and respiratory syndrome virus circulating in Fujian province,a total of 1100 clinical samples(lungs,kidneys,livers,and lymph nodes) and 2846 serum samples were collected from 102 pig farms in different parts of Fujian Province from 2009 to 2017 and these samples were used to detect PRRSV antigen by RT-PCR.Phylogenetic analysis based on the ORF5 gene showed that Fujian PRRSVs are topologically classified in lineages1,3,5.1 and 8.7.Phylogenetic analysis indicated that the viruses in lineage 1 are closely related to NADC30 isolated from America.Lineage 8.7 was predominant strains from 2009 to 2013,but NADC30-like PRRSV had become the predominant strains from 2014 to 2017.Analysis of GP5 of Fujian PRRSVs showed that the isolates had high variability in the primary neutralizing epitope(PNE).Potential N-linked glycosylation sites began to accumulate and were close to the PNE.The above-mentioned results showed that,multiple PRRSV types now co-exist in swine herds,and it is essential to take comprehensive measures to control PRRSV infection in swine herds avoiding introduction of new strains.
