摘要
目的观察高糖状态下原代培养的胸椎黄韧带细胞骨化相关因子骨形态发生蛋白-2(BMP-2)和转化生长因子β(TGF-β)的表达情况,探讨高血糖对胸椎黄韧带细胞骨化的影响。方法使用胸椎黄韧带骨化患者和胸椎骨折患者的黄韧带进行原代培养,分为骨化对照组、骨化高糖组以及骨折对照组、骨折高糖组,在光镜下观察细胞生长状况,免疫细胞化学及Western blot方法检测骨化相关因子BMP-2和TGF-β的表达情况。结果光镜下黄韧带细胞均匀分布,呈长梭形、多角形,胞质丰富,细胞核居中。骨折对照组细胞BMP-2和TGF-β表达量很少,骨化对照组BMP-2和TGF-β的表达明显增加(P<0.05),高糖培养后骨折高糖组及骨化高糖组BMP-2和TGF-β的表达均明显增加(P<0.01),而且骨化高糖组的表达量显著高于骨折高糖组(P<0.01)。结论高血糖促进了胸椎黄韧带细胞骨化相关因子BMP-2和TGF-β的表达,糖尿病/高血糖是胸椎黄韧带骨化的危险因素。
Objective To observe the expression of bone morphogenetic proteins 2( BMP-2) and transforming growth factor-β( TGF-β) in ossification of thoracic ligament flavum primary cells under hyperglycemia condition.Methods Primary thoracic ligament flavum cells from patients with thoracic ossification of ligament flavum and thoracic fractures were cultured and classified four groups: ossification control group,ossification hyperglycemic group and fracture control group,fracture hyperglycemic group. Then the cell growth condition was observed by light microscope and the expression of BMP-2 and TGF-β were detected by immunocytochemistry and Western blot. Results Ligament flavum cells were distributed evenly with long spindle or polygonal shape,and abundant cytoplasm while cell nucleus were in the middle under light microscope. The expression of BMP-2 and TGF-β were significantly increased in ossification control cultures compared with fracture control cells. The expression of ossification relevant factors BMP-2 and TGF-β were increased both in ossification culture cells and fracture culture cells treated with high glucose,and the expression in ossification culture cells was clearly higher than in fracture culture cells.Conclusion Hyperglycemia increase the expression of BMP-2 and TGF-β in thoracic ossification of ligament flavum which is a risk factor in the process of thoracic ossification of ligament flavum.
