高效液相色谱法同时测定植物饮料中7种绿原酸单体含量 被引量:4

Simultaneous Determination of Seven Chlorogenic Acid Monomers in Plant Beverages by High Performance Liquid Chromatography

摘要 建立了同时测定植物饮料中7种绿原酸单体含量的高效液相色谱法。样品经甲醇溶液超声提取,离心后过滤。经十八烷基键合硅胶柱和梯度洗脱程序分离,二极管阵列检测器进行检测。7种绿原酸在给定的范围内呈良好线性,相关系数R^2均大于0.999 3。仪器检出限(LOD)和定量限(LOQ)分别为0.20~0.35 mg/L和0.80~1.20 mg/L。对样品进行三个水平浓度的加标回收实验,回收率在85.8%~95.7%之间,相对标准偏差(RSD)为5.24%~8.82%。说明方法均有操作简便,回收率高,精密度好等优点,适用于植物饮料中7种绿原酸单体含量的日常分析测定。 A high performance liquid chromatography(HPLC) method for simultaneous determination of seven chlorogenic acid monomers in plant beverages was established. The sample was extracted by methanol solution and filtered by centrifugation, and then it was separated and detected by octadecyl bonded silica gel column with gradient elution program and the diode array detector. The seven chlorogenic acids all showed good linearity within a given range, and the correlation coefficient R2 was greater than 0.9993. The instrument detection limit(LOD) and quantitative limit(LOQ) were 0.20-0.35 mg/L and 0.80-1.20 mg/L respectively. The recovery of three levels of the sample was tested. The recoveries were 85.8%-95.7% with relative standard deviations(RSDs) of 5.24%-8.82%. The method is easy to operate, and has high recovery rate, good precision, so it is suitable for the daily analysis and determination of seven kinds of chlorogenic acid monomer in plant beverages.
