摘要
目的:探讨藤黄醇提取物对HCT116细胞侵袭的影响及其内在的分子作用机制。方法:采用Transwell侵袭实验观察藤黄醇提取物对HCT116细胞的侵袭能力影响,实时荧光定量PCR法检测MMP-7和Cyclin D1的mRNA表达水平,Western Blot法检测MMP-7和Cyclin D1的蛋白表达水平,免疫荧光观察β-catenin在细胞内的定位变化。结果:随着藤黄醇提取物剂量的增加,其抑制HCT116细胞的侵袭作用逐渐增强,β-catenin在细胞核内分布逐渐下降,MMP-7和Cyclin D1的mRNA和蛋白表达水平逐渐下降。结论:藤黄醇提取物可能通过抑制异常活化的Wnt经典信号通路来抑制HCT116细胞的侵袭。
Objective: To explore the effect of ethanolic extract of Gamboge on the invasion ability of HCT116 colon cancer cell and underlying molecular mechanism. Methods: The invasion ability of HCT116 colon cancer cell was identified by cell invasion assay. The expressions of mRNA of MMP-7 and Cyclin D1 were examined by real-time fluorescence quantitative PCR analysis. The expressions of proteins were detected by Western Blot analysis. The localization ofβ-catenin was tested by immunofluorescence confocal microscopy. Result: With the dosage of Gamboge increased,the rate of inhibition of HCT116 colon cancer cell was gradually increased and the distribution of β-catenin in nucleus was decresed. Meanwhie,the ethanolic extract of gamboge down-regulated the mRNA and protein levels of MMP-7 and Cyclin D1. Conclusion: The ethanolic extract of Gamboge might inhibit the canonical Wnt signaling pathway to suppress the invasion ability of HCT116 colon cancer cells.
