摘要
目的观察生长抑素(SST)与吉西他滨(Gem)联合应用对人转移性胰腺癌细胞As PC-1增殖的影响。方法体外培养As PC-1细胞。(1)采用0、100、200、400、600、800μg/m L SST处理24、48、72 h,计算SST的半数抑制浓度(IC50);(2)采用0、0.1、1、5、10、20、40、60μmol/L Gem处理48 h(按照SST IC50时间),计算出Gem IC50;(3)依据以上实验得出的SST、Gem IC50,将As PC-1细胞分为4组,A组不做任何处理,B组加入SST IC50,C组加入Gem IC50,D组加入1/2 SST IC50+1/2 Gem IC50,处理时间按SST IC50时间进行,计算各组细胞增殖抑制率。结果 As PC-1细胞在SST浓度为600μg/m L处理48 h时达到半数抑制。As PC-1细胞在Gem浓度为20μmol/L时达到半数抑制。A、B、C、D组细胞增殖抑制率分别为0.00%±3.79%、52.59%±4.57%、48.11%±10.93%、64.86%±4.83%,B、C、D组分别与A组比较,B、C组分别与D组比较,P均<0.05。结论 SST联合Gem可抑制As PC-1细胞的增殖。
Objective To observe the effect of combined therapy with somatostatin (SST) and gemcitabine (Gem) on the proliferation of human metastatic pancreatic cancer AsPC-1 cells. Methods The AsPC-1 cells were cultured in vitro. ① The cells were treated with O, 100,200,400, 600, and 800 μg/mL SST for 24, 48, and 72 h, then we calcu- lated the ICso of SST. ② The cells were treated with 0, O. 1, 1 5, 10, 20, 40, and 60 μmol/L Gem for 48 h, and we cal- culated the ICs0 of Gem. After that, AsPC-1 cells were divided into four groups: an untreated control group (group A), a group treated with SST at IC50 ( group B), a group treated with Gem at ICs0 ( group C), and a group treated with SST + Gem at half of their ICso (group D). At 48 h after treatment, the inhibition rates were calculated. Results For AsPC-1 cells, ICso of SST was 600 tLg/mL at 48 h, and ICs0 of Gem was 20 μmol/L. The inhibition rates in the groups A, B, C and D were 0.00% ±3.79%, 52.59%±4.57%, 48.11% ± 10. 93%, and 64.86% ± 4.83%, respectively. The cell growth was significantly inhibited in the groups B, C, and D as compared with that of group A ( P 〈 0.05 ), and the inhibition rate was significantly higher in the group D than in the groups B and C ( P 〈 0.05 ). Conclusion The combinatioff of SST and Gem can inhibit the growth of AsPC-1 ceils.
