不同月龄SPF级SD大鼠临床生理生化背景数据分析 被引量:4

Analysis of the background data of hemocytology and biochemistry in the blood of SPF Sprague Dawley rats at different ages

摘要 目的收集不同性别、不同年龄阶段SPF级SD大鼠临床生理生化背景数据,观察年龄、性别因素对SPF级SD大鼠血液生理生化指标的影响,探讨数据共享的可行性。方法选取饲养在标准屏障环境中的1~8月龄SPF级SD大鼠100只,雌雄各半,麻醉后腹主动脉采血,血细胞分析仪XT-2000IV、Beckman CX-5全自动生化测定仪及Beckman ACL7000血凝分析仪测定正常大鼠血液的生理生化。结果 1~8月龄SD大鼠的血液生化指标尿素氮(BUN)、磷(P)、钠(Na)、钾(K)、氯(Cl)、钙(Ca)、红细胞分布宽度(RDWSD)、红细胞分布宽度变异系数(RDWCV)、血小板平均体积(MPV)、嗜碱性粒细胞比率(BASO)在性别及年龄之间无统计学意义(P>0.05)。雄性大鼠谷丙转氨酶(ALT)、碱性磷酸酶(ALP)、谷草转氨酶(AST)、肌酸激酶(CK)比雌性偏高(P<0.05),雌性大鼠血糖(Glu)、总蛋白(TP)、白蛋白(ALB)、肌酐(CREA)、甘油三脂(TG)高于雄性(P<0.05);1月龄大鼠的血液学指标WBC、RBC、血红蛋白量(HGB)、红细胞压积(HCT)、红细胞平均体积(MCV)、红细胞平均血红蛋白含量(MCH)、红细胞平均血红蛋白浓度(MCHC)、RDWSD、网织红细胞比率(RET)雌雄差异有统计学意义(P<0.05),2月龄大鼠WBC、MONO、RET雌雄差异有统计学意义(P<0.05),3月龄大鼠WBC、RBC、HGB、PLT、NEUT、EO离子含量雌雄差异有统计学意义(P<0.05),4~5月龄大鼠WBC、PLT、PCT、NEUT、MONO、EO雌雄差异有统计学意义(P<0.05),7~8月龄大鼠WBC、NEUT、MONO、RET雌雄差异有统计学意义(P<0.05);1~8月龄大鼠的血凝指标PT、APTT值在性别及月龄之间差异无统计学意义(P>0.05),FBG值随月龄增加先升高而后降低(P<0.05),TT值随月龄增加逐渐升高后保持平稳且差异有统计学意义(P<0.05)。结论 SD大鼠部分的血液生理生化指标在不同月龄、性别之间保持稳定,无差异性;部分指标存在月龄、性别差异且具有规律性可循,有数据共享的可行性。 Objective To establish normal range and mean values of serum biochemical measurements in SPF SD rats with different age and sex used in data sharing. Methods 50 male and 50 female SPF SD rats were assigned into seven groups according to age and subject to blood physiological and biochemical indicator detection by XT-2000IV Vet Auto Hematology Analyzer, Beckman CX-5 automatic biochemistry analyzer and Beckman ACL7000 automatic blood coagulation analyzer. Results The values of the blood physiological and biochemical indicators of all rats, including blood urine nitrogen (BUN), P, Ca, Na, K, C1, red blood cell distribution width (RDWSD) , red blood cell distribution width-correlation variance (RDWCV), mean platelet volume (MPV) and basophil (BASO) were surprisingly consistent (P〉O.05). The levels of alanine aminotransferase (ALT) , alkaline phosphatase (ALP) , aspartate aminotransferase (AST) and creatine kinase (CK) in the blood of male were higher than female, meanwhile, the levels of (Glu) , total protein (TP) , albumin (ALB) , creatinine (CREA) and triglyceride (TG) in the blood of female were higher than those in male.The differences of WBC, RBC, hemoglobin (HGB) , hematocrit (HCT) , mean corpuscular volume (MCV) , mean corpuscular hemoglobin (MCH) , mean corpuscular hemoglobin concentration (MCHC) , RDWSD and reticulocyte (RET) were significant between the female and male rats of 1 month old (P〈0.05). The differences of WBC, MONO and RET were significant in the 2 months old (P〈0.05). The differences of WBC, RBC, HGB, PLT, NEUT and EO were significant between the female and male rats of 3 month old (P〈0.05). The differences of WBC, PLT, PCT, NEUT, MONO and EO were significant between the female and male rats of 4-5 month old (P〈0.05). The differences of WBC, NEUT, MONO and RET were significant between the female and male rats of 7-8 month old (P〈0.05). The coagulation data of PT and APTT in all rats were almost consistent (P〉0.05), while the level of FBG in the blood of all rats increased with age and then decreased, and TT in all rats increased with age and then keeping smoothly as the age of rats increased (P〈0.05). Conduslorl Some markers of hemocytology and biochemistry were the same between male and female SPF SD rats at different ages, however some markers were different but with regularity.
