摘要
以霞晖6号春季嫩梢为外植体材料,通过消毒获得无菌苗,比较不同基本培养基及激素种类浓度对桃增殖、生根的影响,并筛选叶片愈伤诱导条件。结果表明:最适增殖培养基为MS+6-BA 2.0 mg/L+IBA 0.5 mg/L+2.0%蔗糖+5.0 g/L琼脂粉,其平均增殖倍数可达5.2棵/月;适宜生根培养基为MS(大量元素减半)+IAA 2.0 mg/L+2.0%蔗糖+5.0g/L琼脂粉,直接光照培养生根;叶片愈伤诱导最佳培养基为MS+TDZ 5.0 mg/L+IBA 0.2 mg/L+2,4-D 2.0 mg/L+2.0%蔗糖+5.0 g/L琼脂粉,其诱导率可达100%;加入PVP具有较好的抗氧化作用。
The young stems of peach variety Xiahui 6 in spring were used as explants,and the effects of various basic culture media,hormone kinds and concentration on the leaf callus induction,bud proliferation and rooting were studied. The results showed that the optimal medium for bud proliferation was MS + 6-BA 2. 0 mg/L + IBA 0. 5 mg/L + 2. 0% sucrose + 5. 0 g/L agar powder,in which the plantlet was stronger and the average proliferation rate reached 5.2 times per month. The optimal medium for rooting was MS(the content of major elements was halved) + IAA 2.0 mg/L+ 2.0% sucrose + 5.0 g/L agar powder; the direct sunlight was suitable for the rooting of plantlets. The best medium for callus induction of leaf was MS+TDZ 5.0 mg/L+IBA 0.2 mg/L+2,4-D 2.0 mg/L+ 2.0% sucrose+ 5.0 g/L agar powder,and the induction rate could reach 100%; the PVP added into culture medium had a good antioxidant effect.
