南非蜱虫套式PCR鉴定方法的建立

摘要 为探讨来自南非不同蜱种的系统进化关系,建立南非截获蜱虫的套式PCR鉴定方法。根据GenBank中蜱虫的18SrRNA基因序列,设计2对特异性引物,通过优化条件建立蜱虫套式PCR鉴定方法。从南非进口生牛皮上截获寄生蜱,经过形态学初步鉴定后,应用所建立的套式PCR扩增获得10种南非蜱虫(T1-T10)的18S rRNA基因序列,测序后与NCBI中已公布的蜱虫18S rRNA基因序列进行同源性分析。应用MEGA5.0软件进行系统进化分析发现,T1、T2、T3、T4、T7、T8是花蜱属的亚种,T5和T6分别是牛蜱属的不同种或亚种;T9和T10是扇头蜱属的不同种或亚种。说明建立的套式PCR检测方法能够在传统形态学分类的基础上,准确鉴定不同形态的蜱种。 To investigate the phylogenetic relationship of different tick species from South Africa, nested-PCR was established for the intercepted ticks. According to the sequences of 18 S rRNA genes of ticks,twopairs of specific primers were designed, the conditions were optimized and the nested-PCR assay for detec-tion of ticks was established. Ticks were intercepted from raw cowhides imported from South Africa, andafter preliminary morphological identification,the 18 S rRNA gene sequences of 10 kinds of ticks (T1-T10)were obtained by Nested-PCR and subsequently sequenced. Homology analysis of 18 S rRNA was done be-tween the collected ticks and the published ticks in NCBI. Phylogenetic analysis by MEGA5.0 showed thatT1 ,T2 ,T3 ,T4 ,T7 and T8 were subspecies of Amblyornma ,T5 and T6 were different species or subspeciesof Boophilus,T9 and T10 were different species or subspecies of Rhipicephalus. The nested-PCR estab-lished in this study can accurately identify different species of ticks on the basis of traditional morphologicalidentification.
