摘要
目的观察补肾益髓方及其拆方补肾方与化痰活血方对星形胶质细胞损伤后细胞活性的影响及胶质瘢痕中胶质纤维酸性蛋白(GFAP)、硫酸软骨素糖蛋白(CSPGs)表达的影响。方法体外原代培养星形胶质细胞,划痕法建立胶质瘢痕模型,分别加入补肾益髓方、补肾方和化痰活血方含药血清,继续培养24 h后,用CCK-8法测定细胞存活率,高内涵法测定GFAP与CSPGs蛋白的表达。结果与正常对照组比较,模型组细胞存活率明显下降(P<0.01),GFAP与CSPGs蛋白显著上调(P<0.01)。与模型组相比,补肾益髓方、补肾方及化痰活血方含药血清均能够促进星形胶质细胞细胞存活率(P<0.01);均能够下调CSPGs蛋白的表达(P<0.01或P<0.05)。补肾益髓方及补肾方升高细胞存活率优于化痰活血方(P<0.05或P<0.01);降低CSPGs蛋白的表达方面补肾益髓方优于化痰活血方(P<0.05)。结论补肾益髓方及其拆方均能够抑制损伤后星形胶质细胞胶质瘢痕中CSPGs蛋白的表达,以促进轴突的修复与再生,以补肾益髓方效果最佳。
Objective: To observe effects of Bushen Yisui Decoction (BSYS) and its disassembled formulas, Bushen Decoction(BS) and Huatan Huoxue Decoction(HTHX) on the cell viability of impaired astmcytes and on the expression of glial fibrillary acidic protein(GFAP) and chondrotin sulphate proteoglycans(CSPGs) in astrocyte with glial scar. Methods: The astroeytes were cultured and the astrocyte scratch injury model was established in vitro,and then the cells were treated with serum containing medicine of BSYS Decoction,BS Decoction and HTHX Decoction for additional 24 h. The cell viability was measured with the method of cell counting Kit(CCK) -8;the expression of GFAP and CSPGs were measured with high throughput screening(HTS). Results: Compared with the normal control group (NC),the cell viability of the model group (MO) was significantly decreased (P〈 0.01 ). Meanwhile,the expression of GFAP and CSPGs significantly increased (P〈 0.01). Compared with the MO group ,the serum containing BSYS Decoction,BS Decoction and HTHX Decoction significantly increased the cell viability (P〈0.01),and significantly decreased expression of CSPGs (P〈 0.01 or P〈 0.05). The effect of serum containing BSYS Decoction,BS Decoction on the cell viability was better than that of HTHX Decoction (P〈 0.01 or P〈 0.05) and the effect of BSYS Decoction on decrease of CSPGs expression was better than HTHX Decoction (P 〈 0.05). Conclusion: BSYS Decoction,BS Decoction and HTHX Decoction can decrease the expression of CSPGs in glial scar, and promote the regeneration and repair of axon, especially in BSYS Decoction.
