摘要
基因组编辑技术是指在基因组的特定位点,进行基因序列修改的技术。随着基因组编辑工具的研究的深入,人工核酸酶技术和RNA介导的CRISPR/Cas9系统成为两类应用广泛的工具。后者具备构造简单且效率高的优势,使其迅速发展成为应用最为广泛的基因组编辑工具。肿瘤的治疗由于其复杂的病因和肿瘤细胞对于免疫过程的逃逸而进展缓慢,T细胞的武装技术为肿瘤的免疫治疗提供了新的思路。本文综述了CRISPR/Cas9系统的原理、特点以及其在肿瘤免疫治疗中的应用。
Genome editing technology refers to the gene sequence modification technology in specific site of the genome. With the deeper study of genomic editing tools, the artificial nuclease technology and RNA-mediated CRISPR/Cas9 system have become two widely used tools. The latter has the advantage of simple structure and high efficiency, which makes it become the most widely used tool for genome editing. The treatment of tumor progresses slowly due to its complicated etiology and the escape of tumor cells from immune process. The T cell armed technology provides a new idea for tumor immunotherapy. The principle, characteristics and application of CRISPR/Cas9 system in tumor immunotherapy were reviewed.
