摘要
目的本实验采用Agilent 1220高效液相色谱法建立黄藤的指纹图谱,并建立了同时测定盐酸巴马汀、盐酸药根碱含量的方法。方法采用ACE C_(18)(4.6mm×250mm,5μm)色谱柱,0.1%磷酸溶液(每100mL中加入0.1g十二烷基硫酸钠)(A)-乙腈(B)为流动相梯度洗脱,流速1.0m L/min,柱温30℃,检测波长345nm。结果建立了黄藤的HPLC定量指纹图谱,10批药材的HPLC指纹图谱共标定12个共有峰,相似度均在0.90以上。结论该方法建立的黄藤HPLC指纹图谱特征性强、重复性好、灵敏度高,为黄藤的鉴定及质量控制提供了参考价值。
Objective The experiment adopted Agilent 1220 HPLC to establish the fingerprint of Fibraurea recisa Pierre, and establish the way that simultaneously determined the content of palmatine hydrochloride and jatrorrhizine hydrochloride. Methods The analytical column was ACE C_(18)(4.6 mm×250 mm,5μm).The mobile phase was acetonitrile-water(containing 0.1% phosphoric acid and 0.1% sodium dodecyl sulfate) with gradient elution,and the detection wavelength was set at 345 nm.The flow rate was 1.0 m L/min,and the column temperature was set at 30 ℃. Results The HPLC ration fingerprint of Fibraurea recisa Pierre was established.12 common peaks were defined among the 10 batches in the HPLC fingerprint, and the similarity is above 0.90. Conclusion The fingerprint of Fibraurea recisa Pierre that the method has strong features, repeatability and high sensitivity,which provides reference value for the identification and the quality control of Fibraurea recisa Pierre.
