丙泊酚对离体大鼠心脏代谢性酸中毒的影响 被引量:3

摘要 目的观察丙泊酚对离体大鼠心脏代谢性酸中毒的影响。方法将24只大鼠采用随机数字法分为3组:对照组(PC组),25μmol/L丙泊酚组(PL组),50μmol/L丙泊酚组(PH组),每组8只。PC组先用常规K-H液(pH 7.4)灌注40 min,然后用酸性K-H液(pH 6.5)灌注20 min,诱发心脏整体的代谢性酸中毒,最后用常规K-H(pH 7.4)液恢复灌注30 min;PL组和PH组的灌注流程与PC组相同,差别是平衡灌注25 min后向K-H液中分别加入相应浓度的丙泊酚。记录心电图,检测肌酸激酶(CK)、乳酸脱氢酶(LDH)含量。测定左心室部分心肌组织丙二醛(MDA)含量和超氧化物歧化酶(SOD)活力。结果 PC组和PL组的心律失常时间分别为(126.9±15.4)s、(117.6±8.2)s,均长于PH组[(33.6±6.4)s](P<0.05)。PL组和PH组的CK和LDH含量均低于PC组(P<0.05)。PL组和PH组的MAD含量分别为(3.3±0.5)nmol/mgprot、(3.4±0.4)nmol/mgprot,均低于PC组的(6.7±0.8)nmol/mgprot(P<0.05)。PL组和PH组的SOD活性分别为(33.6±4.3)U/mgprot、(32.4±3.5)U/mgprot,均高于PC组的(25.5±4.2)U/mgprot(P<0.05)。结论适当浓度的丙泊酚对离体大鼠心脏代谢性酸中毒有一定的保护作用,在本研究的实验条件下,丙泊酚在抗心律失常、改善心脏电生理方面,50μmol/L丙泊酚优于25μmol/L丙泊酚。在降低心肌酶含量和抗脂质过氧化上,50μmol/L和25μmol/L丙泊酚均具有明显的作用。 Objective To study effects of propofol on metabolic acidosis in isolated rat hearts. Methods 24 SD rats,male and adult,were divided into 3 groups( 8 rats in each group) using a random number: a control group( PC group),a25 μmol/L propofol group( PL group) and a 50 μmol/L propofol group( PH group). The isolated hearts in PC group were perfused with K-H solution( pH 7. 4) for 40 min,then were subjected to acidic K-H solution( pH 6. 5) for 20 min,and followed to restore perfusion with the K-H solution( pH 7. 4) for 30 min. Perfusion process of PL group or PH group was similar to PC group,but 25 μmol/L or 50 μmol/L propofol was added to K-H solution at 25 min after stable perfusion,respectively. The time of arrhythmias was recorded and calculated. The contents of creatine kinase( CK) and lactate dehydrogenase( LDH) in the effluent were measured. The myocardial tissue of left ventricle was obtained after reperfusion to detect MDA and SOD activity. Results The arrhythmias time of PC group,PL group and PH group respectively were( 126. 9 ±15. 4) s,( 117. 6 ± 8. 2) s and( 33. 6 ± 6. 4) s. The time of arrhythmias of the PC group and PL group was more than that of the PH group( P 0. 05). The activities of CK and LDH in PL group and PH group were lower than those in the PC group( P 0. 05). The MAD of PC group,PL group and PH group respectively were( 6. 7 ± 0. 8) nmol/mgprot,( 3. 3 ±0. 5) nmol/mgprot and( 3. 4 ± 0. 4) nmol/mgprot,PL and PH group' s level was significantly lower than that of the PC group( P 0. 05). SOD level of the PC group,PL group and PH group respectively were( 25. 5 ± 4. 2) U/mgprot,( 33. 6 ±4. 3) U/mgprot and( 32. 4 ± 3. 5) U/mgprot,PL and PH group' s level was significantly higher than that of the PC group( P 0. 05). Conclusion Appropriate concentration of propofol can protect isolated rat heart against metabolic acidosis.Under the present experimental conditions,50 μmol/L propofol has better reduced arrhythmia and improved cardiac electrophysiology on isolated rat heart subjected to metabolic acidosis than 25 μmol/L propofol. On reducing myocardial enzymes and preventing lipid peroxidation,25 and 50 μmol/L propofol both have significant function.
