摘要
目的对系统性红斑狼疮小鼠白细胞介素22(IL-22)细胞因子的表达情况进行研究,探讨IL-22在系统性红斑狼疮发病中的意义。方法将80只小鼠随机分为系统性红斑狼疮组和对照组,每组各40只。系统性红斑狼疮组小鼠经尾静脉注射活化的脾细胞复制系统性红斑狼疮模型,对照组小鼠经尾静脉注射等量的生理盐水。比较模型复制后第4周和第8周时两组小鼠肾脏HE染色、血清抗双链DNA抗体滴度、血清IL-22水平、肾脏组织和淋巴组织IL-22 m RNA表达、肾脏和淋巴结组织中IL-22蛋白的表达。结果系统性红斑狼疮组小鼠肾小管上皮细胞变性,肾间质内见大量炎症细胞浸润,肾小球系膜细胞增生,对照组小鼠肾脏组织形态正常。系统性红斑狼疮组小鼠第4周和第8周时血清抗双链DNA抗体滴度与对照组比较,差异有统计学意义(P<0.05),系统性红斑狼疮组高于对照组。系统性红斑狼疮组复制模型第4周和第8周时小鼠血清IL-22和对照组比较,差异无统计学意义(P>0.05);肾脏组织IL-22 m RNA水平与对照组比较,差异有统计学意义(P<0.05);淋巴组织IL-22 m RNA水平和对照组比较,差异无统计学意义(P>0.05)。IL-22蛋白在系统性红斑狼疮小鼠肾脏中呈强阳性表达,在肾小球中比较集中,在对照组小鼠肾脏中表达不明显。IL-22蛋白在两组小鼠淋巴组织中均没有表达。结论系统性红斑狼疮小鼠肾脏出现系统性红斑狼疮肾的病理学改变,肾脏组织中IL-22 m RNA和IL-22蛋白的表达升高。
Objective To study the expression of interleukin-22(IL-22) in mice with systemic lupus erythematosus(SLE), and explore the significance of IL-22 in the pathogenesis of SLE. Methods Eighty mice were randomly divided into SLE group(40 cases) and control group(40 cases). The mice of the SLE group were injected with activated splenocytes through caudal vein to establish SLE mouse model. Those of the control group were injected with the same amount of normal saline through caudal vein. In the 4 th and 8 th weeks after model establishment, the kidney morphology on HE sections, the titer of serum anti-double stranded DNA antibody, serum IL-22 level, the expressions of IL-22 m RNA and protein in the kidney and lymphoid tissues were compared between the two groups.Results Histological study revealed degeneration of renal tubular epithelial cells, a large number of inflammatory cell infiltration in the renal interstitium, and proliferation of mesangial cells in the SLE group; but normal renal morphology in the control group. In the 4 th and 8 th weeks after modeling, the serum titer of anti-double stranded DNA antibody in the SLE group was higher than that in the control group(P < 0.05); the serum level of IL-22 in the SLE group was not statistically different from that in the control group(P > 0.05); the expression of IL-22 m RNA in the kidney tissues of the SLE group was higher than that of the control group(P < 0.05); but the expression of IL-22 m RNA in the lymphoid tissues of the SLE group was not statistically different from that of the control group(P >0.05). The expression of IL-22 protein in the kidneys of the SLE group was strongly positive and concentrated in the glomeruli, while IL-22 protein was not obviously expressed in the kidneys of the control group. IL-22 protein was not expressed in the lymphoid tissues of either group. Conclusions The systemic lupus erythematosus mice have the pathological changes of systemic lupus erythematosus in the kidneys. The expressions of IL-22 m RNA and protein are increased in the renal tissues.
