摘要
本试验采用PCR-SSCP和PCR-RFLP方法分析了黑素皮质素受体4(MC4R)基因在5个地方鸡种中的遗传多态性,结果在5个供试鸡群体中检测到两个突变位点。在NC_006089.4:g.54 C>G位点中,CC基因型在莱芜黑鸡和琅琊鸡中均没有检测到,CG杂合型在汶上芦花鸡中表现为优势基因型,突变型GG基因型在其余4个群体中表现为优势基因型,等位基因G在所有供试群体中表现为优势等位基因。在NC_006089.4:g.315位点中,GT杂合型在汶上芦花鸡中表现为优势基因型,GG野生型在其余4个群体中表现为优势基因型,等位基因G在除汶上芦花鸡以外的4个群体中表现为优势等位基因。
The SNPs of MC4R gene in five Chinese indigenous Chicken Breeds were detected by PCR-SSCP and PCR-RFLP methods. As a result,two mutations in MC4R gene were identified. In NC006089.4: g. 54 C G,genotype CC was not found in Laiwu Black chickens and Langya chickens,the frequency of genotype CG was higher than that of the other genotypes in Wenshang Barred chickens,and GG showed to be the advantageous genotype in the other four populations; allele G was dominant in all the five populations. In NC006089. 4: g. 315,heterozygous genotype GT was advantageous in Wenshang Barred chickens,and GG was advantageous in other four populations; allele G was dominant in the four populations except Wenshang Barred chickens.
