摘要
核酸、蛋白质、糖类是生命系统中的主要大分子物质。随着生物化学和分子生物学的发展,科学家们对核酸和蛋白质的研究逐渐深入,对其在决定生命规律方面的作用已有了初步的了解。但相比之下对生物多糖的研究较滞后,尽管糖类的生理意义同样至关重要。在生物体中生物多糖(polysaccharide)多是以糖缀合物(glycoconjugates,即多糖与非糖物质如脂类或蛋白质共价结合而成的糖复合物)的形式存在。
Carbohydrates are important structural and functional moieties of glycoproteins and glycolipids . Many important biological processes, such as development, immunology, cancer metastasis,etc. are mediated by the carbohydrates moieties of glycoconjugates. Oligosaccharides from most biological sources are of considerable complexity and variety. Understanding the precise roles they play in biological processes often required their isolation, detailed structural analyses and usage in functional assays. Generally oligosaccharides lack chromophore groups, which increase the difficulty of oligosaccharides structure analysis. Now a complete analysis of a carbohydrate oligomer is one of the foremost and compelling challenges in structural biology. We reviewed 28 references on the methods for isolation and structure determination for oligosaccharides.
