摘要
应用重组聚合酶扩增技术(recombinase polymerase amplification,RPA),根据玛卡特异性基因defensin序列设计筛选了1对可用于扩增的引物,建立了玛卡及其加工产品特异性源性成分的RPA检测方法。该方法具有较高的灵敏性,检测灵敏度达到0.1 ng/L以上,操作方便,可以在室温条件下完成反应,检测可在0.5 h内完成,极大缩短了检测时间,适用于市场上玛卡及其制品的快速鉴别。
According to the defensin gene sequence of Lepich'um meyenii, a pair of RPA primers was designed and a event-specific detection method for Lepich'um meyenii and its derivates based on recombinase ploymerase amplification (RPA) was established. This method had high sensitivity with 0.1 ng/pL of DNA sensitivity, and was simple to operate. It could complete reaction at room temperature. The process could finish detection in 0.5 h. Detection time was greatly shortened. The RPA method was suitable for rapid identification ofLepidium meyenii and its derivates from the market.
