摘要
目的探讨MicroRNA(miR-182)在宫颈癌组织、宫颈上皮内瘤变组织(CIN)和正常宫颈组织中的表达,及其对宫颈癌细胞增殖、侵袭和迁移能力的影响。方法应用实时聚合酶链反应(real-time PCR)技术方法检测78例宫颈癌组织、30例CIN组织及20例正常宫颈组织中miR-182的表达,并分析其与宫颈癌临床病理特征之间的关系。在宫颈癌细胞株CaSki中,应用细胞转染技术上调miR-182的表达,应用Cell Counting Kit-8(CCK-8)试剂盒检测细胞增殖能力的变化,Transwell实验检测细胞侵袭迁移能力的变化。结果 Realtime PCR结果显示miR-182在宫颈癌组织、宫颈上皮内瘤变(CIN)组织与正常宫颈组织比较,差异有统计学意义﹙P<0.05﹚;两两比较,miR-182在宫颈癌组织和CIN组织中的表达较正常宫颈组织降低﹙P<0.05﹚,miR-182在宫颈癌组织与CIN组织比较,差异无统计学意义(P>0.05),宫颈癌组织中miR-182的表达与分化程度、淋巴转移密切相关,而与年龄、肿瘤直径、浸润深度及临床分期等无关(P>0.05)。在人宫颈癌上皮细胞(CaSki)中,上调miR-182表达能够抑制肿瘤细胞的侵袭和迁移﹙P<0.05﹚,但不改变细胞的增殖能力。结论 miR-182在宫颈癌组织中低表达,miR-182的表达水平与分化程度和淋巴结转移有关,上调miR-182的表达能够抑制宫颈癌CaSki细胞的迁移及侵袭。
Objective To invest igate the expression of miR-182 in cervical cancer , cervical int raepi thel ial neoplasia (CIN) and normal cervical tissue, and to discuss its ef fect on proli ferat ion, invasion and migrat ion of cervical cancer cells. Methods The expression level of miR-182 was detected by real -t ime PCR in 78 samples of cervical cancer tissues, 30 samples of CIN tissues and 20 samples of normal cervical t issues. The relat ionships between miR-182 expression and cl inicopathological features of cervical cancer were analyzed. Cervical cancer CaSki cel ls were transfected wi th miR-182 mimics. CCK-8 assay and Transwel l assay were used to analyze the effects of miR-182 on the prol i ferat ion, invasion and migrat ion of cervical cancer cel ls. Resul ts The expression level of miR-182 showed signi f icant dif ferences among the cervical cancer t issues, the CIN tissues and the normal cervical tissues (P〈0.05). Pairwise compar isons showed the miR-182 expression in the cervical cancer tissues and the CIN tissues was signi ficantly lower than that in the normal cervical tissues (P 〈0 .05 ) , but the dif ference between the cervical cancer tissues and the CIN tissues was not stat ist ical ly signi f icant (P〉 0.05) . The expression level of miR-182 in the cervical cancer tissues was signi ficantly correlated wi th degreeof di fferentiat ion and lymphat ic metastasis(P〈0.05) , but not in signi ficant correlat ion wi th age, tumor diameter , depth of invasion or cl inical stages (P〉0.05) . Transwel lassay showed that up-regulated miR-182 expression would signi f icantly reduce the migrat ion and invasion of the cervical cancer CaSki cel ls (P〈 0.05). CCK-8 assay showed that up-regulated miR-182 expression had no ef fect on the prol i ferat ion of the cervical cancer cel ls (P〉0.05). Conclusions The expression level of miR-182 is obviously decl ined in cervical cancer tissues and is related to degree of dif ferentiat ion and lymphat ic metastasis. The up-regulated miR-182 expression can inhibi t the invasion and migrat ion of cervical cancer CaSki cells.
