摘要
对超声辅助混合酶法提取米糠水溶性膳食纤维(RSDF)的提取工艺进行了研究。考察了酶及其添加量、料液比、超声功率和超声时间等对RSDF得率的影响。经过单因素试验和central composite design中心复合响应面分析法,确定了RSDF的最佳提取工艺为:添加6%的木瓜蛋白酶,60℃水解1h并灭酶后,再添加1.5%木聚糖酶、料液比1∶25g/ml,55℃水浴超声(600 W)5min。在此条件下,米糠水溶性膳食纤维得率最高为25.10%。
The aim was to optimize the extraction process of rice bran soluble dietary fiber (RSDF) by ultrasonic assisted mixed enzyme methods. These factors which influence the extraction efficiency were discussed: the added amount of enzyme, solid to liquid ratio, ultrasonic time and power, water bath temperature and extraction time. The single factor and CCD of re-sponse surface analysis showed that the optimum extraction conditions were: papain amount 6% , hydrolysis of 1 h at 60℃ for enzyme inactivation,then adding xylanase dosage 1. 5% , solid to liquid ratio of 1 : 25 g/ml,ultrasonic time (600 W) of 5 min at 55℃. Under this conditions, the yield of RSDF was up to 25. 10%.
