高脂饮食对ApoE^-/-小鼠血浆及主动脉视黄醇结合蛋白4表达的影响及意义 被引量:1

摘要 目的观察高脂饮食对apoE^(-/-)小鼠血浆及主动脉RBP4水平的影响,探讨其意义。方法 8周龄C57 BL/6及apoE^(-/-)小鼠分为4组:C57 BL/6小鼠普食组,C57 BL/6小鼠高脂组,apoE^(-/-)小鼠普食组,apoE^(-/-)小鼠高脂组。喂养不同周数(0,1,2,3,4周)后取血浆用于脂质和血糖的测定,应用免疫印迹法检测血浆及附睾脂肪组织RBP4的表达,免疫组化检测主动脉RBP4的表达。结果与C57BL/6小鼠普食组及apoE^(-/-)小鼠普食组相比,喂养4周apoE^(-/-)高脂组小鼠血浆胆固醇(TC)、甘油三酯(TG)及血糖(GLU)升高(P<0.05),其血浆及脂肪组织RBP4的表达水平也升高(P<0.05)。apoE^(-/-)高脂组小鼠的主动脉RBP4的免疫阳性表达较C57BL/6高脂组小鼠及apoE^(-/-)普食组小鼠增加,且主动脉粥样硬化斑块内可见到明显的RBP4表达。结论高脂饮食诱导apoE^(-/-)小鼠血浆、脂肪组织及主动脉RBP4表达升高,RBP4可能参与了代谢综合征,特别是血管病变的病理过程。 Objective To explore the effect of high fat diet on RBP4 expression in plasma and aorta of apoE^-/-mice. Methods The8-week-old C57BL/6 and apoE^-/-mice were divided into four groups: C57BL/6 control group(fed with normal diet) and C57BL/6high-fat diet group(C57 HF),apoE^-/-control group and apoE^-/-high-fat diet group(apoE^-/-HF). Plasma lipids were detected after different feeding time by enzymic colorimetric method. Glucose was determined using glucose oxidase method. The protein levels of RBP4 in plasma and epididymal adipose tissue were analyzed by Western blot. RBP4 expression of aorta was determined using immunohistochemistry. Results In comparison with C57 mice and apoE^-/-control mice,plasma levels of total cholesterol(TC),triglyceride(TG) and glucose(GLU) significantly increased after high-fat diet for 4 weeks in apoE^-/-HF mice(P〈0. 05),and the expression of RBP4 in plasma and adipose tissue was also increased in apoE^-/-HF mice(P〈0. 05). In comparison with C57 HF mice and apoE^-/-control mice,RBP4 immunopositive blots were obviously increased in the aorta of apoE^-/-HF mice,with strong positive staining in atherosclerotic lesions. Conclusion High-fat diet increases RBP4 expression in plasma,adipose tissue and aorta of apoE^-/-mice,suggesting that RBP4 may participate in pathology of metabolic syndrome,especially the vascular disease.
