摘要
目的:观察厚叶五味子对大鼠佐剂性关节炎(AA)的治疗作用,并从细胞因子水平探讨其作用机制。方法:采用完全弗氏佐剂(CFA)制备大鼠AA模型,通过关节肿胀度、关节炎评分、免疫器官指数、苏木精-伊红(HE)染色等指标评价厚叶五味子对AA大鼠的治疗作用;利用酶联免疫吸附试验(ELISA)法检测AA大鼠血清中TNF-α、IL-6和IL-1β。结果:与模型组相比,厚叶五味子总浸膏(KCTE)对AA大鼠原发性和继发性足跖肿胀的抑制作用显著,在下调大鼠整体关节炎评分、免疫器官系数,改善大鼠踝关节病理状态等方面同样具有显著作用,能够显著降低大鼠血清中TNF-α和IL-6的含量,并呈现出一定的量效关系。结论:KCTE对大鼠AA的治疗作用显著,其机制可能与调节机体免疫功能、减少炎性细胞因子TNF-α和IL-6的分泌有关。
Objective: To investigate the therapeutic effect of Kadsura coccinea on adjuvant arthritis( AA) rats and its mechanism from cytokine levels. Methods: The complete Freund's adjuvant( CFA)-induced AA rats model was used.The therapeutic effect of K. coccinea on AA rats was evaluated by the articular swelling,arthritis score,immune organ index and hematoxylin-eosin( HE) staining. The inflammatory factors of TNF-α,IL-6,and IL-1β in AA rat serum were detected by enzyme-linked immunosorbent assay( ELISA). Results: Compared with the model group,the total extract of K. coccinea( KCTE) could significantly inhibit the primary and secondary articular swelling,lower the arthritis score and immune organ index and improve the ankle pathologies in AA rats. The high and middle dose groups( KCTE-H and KCTE-M) could significantly reduce the serum levels of TNF-α and IL-6,exhibiting a certain dose-effect relationship. Conclusion: KCTE showed significant therapeutic effect on AA rats and the mechanism may be associated with modulation of immune function and down-regulation of inflammatory cytokines TNF-α and IL-6.
