人参三七川芎醇提物对衰老人心脏微血管内皮细胞自噬的影响 被引量:12

摘要 目的观察人参三七川芎醇提物延缓内皮细胞衰老的可能作用机制。方法实验分为衰老细胞组、自噬模型组、白藜芦醇组、人参三七川芎醇提物组,除衰老细胞组外,其余各组的衰老人心脏微血管内皮细胞(HCMEC)通过饥饿方法建立自噬模型。白藜芦醇组用10μmol/L白藜芦醇溶液培养,人参三七川芎醇提物组用100 mg/L人参三七川芎提取物溶液培养48 h。采用电子显微镜观察细胞中自噬小体的形态学改变,western blot法检测自噬蛋白Beclin1、LC3B表达情况。结果与自噬模型组比较,人参三七川芎醇提物组和白藜芦醇组细胞质中出现较多的独立双层及多层膜结构的自噬体。与自噬模型组比较,人参三七川芎醇提物组和白藜芦醇组Beclin1、LC3B蛋白表达明显增多(P<0.01),且白藜芦醇组Beclin1、LC3B蛋白的表达明显多于人参三七川芎醇提物组(P<0.01)。结论人参三七川芎醇提物可促进衰老内皮细胞自噬体的产生及自噬蛋白的表达,这可能是其延缓内皮衰老的重要机制之一。 Objective To observe the possible mechanism about alcohol extract of Renshen [ Radix Ginseng, 人参 ], Sanqi [ Radix Notoginseng, 三七 ] and Chuanxiong [ Rhizoma Ligustici Chuanxiong, 川芎 ] in postponing endo- thelial cells senescence. Methods The experimental subjects were divided into the senile ceils group, the model of autophagy group, the resveratrol group and the alcohol extract of Renshen, Sanqi and Chuanxiong group. Except for the senile ceils group, human senescent cardiac microvascular endothelial cells were made into model of autophagy by hunger in the rest of the groups. The resveratrol group was cultivated with 10 μmol/L of resveratrol solution, and the alcohol extract of Renshen, Sanqi and Chuanxiong group was cultivated with 100 mg/L of alcohol extract solution of Renshen, Sanqi and Chuanxiong, both for 48 h. Morphological changes of autophagosome in the cells were observed by using electron microscope, and expressions of autophagy protein Beclinl and LC3B were detected with western blot method. Results Compared with the model of autophagy group, there were more autophagosome with independent double and multilayer membrane structure in cytoplasm in the resveratrol group and the alcohol extract of Renshen, Sanqi and Chuanxiong group. Compared with the model of autophagy group, the expression of Beclinl and LC3B significantly increased in the resveratrol group and the alcohol extract of Renshen, Sanqi and Chuanxiong group (P 〈 0. 01 ). Compared with the alcohol extracts of Renshen, Sanqi and Chuanxiong group, the expression of Beclinl and LC3B significantly increased in the resveratrol group (P 〈 0. 01 ). Conclusion The alcohol extract of Renshen, Sanqi and Chuanxiong may promote autophagosome to generate in senescent endothelial cells and autophagy protein to express. This may be the one of the important mechanisms about postponing endothelial senescence
