摘要
目的:比较自然衰老法与卵巢去势法建立的ICR小鼠围绝经期模型,并对模型进行评价。方法:通过观察阴道脱落细胞,比较自然衰老组及卵巢去势组小鼠的动情周期情况;观察小鼠悬尾活动,比较自然衰老组及卵巢去势组小鼠的情绪状态;测定血清性激素,比较血清卵泡刺激素(FSH)、促黄体激素(LH)、雌二醇(E2)的含量;测定小鼠下丘脑神经肽,比较下丘脑Kiss-1及其受体GPR54 mRNA的表达。结果:自然衰老组及卵巢去势组小鼠动情周期均紊乱,动情频率均减少;两组小鼠悬尾活动比均显著提高;两组小鼠血清的FSH、LH水平显著上升,E2水平明显降低;两组小鼠的下丘脑Kiss-1及受体GPR54 mRNA的表达显著升高。相比卵巢去势小鼠,自然衰老小鼠动情周期相对延长,动情频率明显减少,血清的FSH、LH水平上升更显著,E2水平降低更显著,Ki SS-1/GPR54 mRNA的表达升高更明显。结论:自然衰老法与卵巢去势法均能建立小鼠围绝经期模型,而自然衰老法优于卵巢去势法。
Objective：A comparative study on the establishment of the model of perimenopausal syndrome by natural aging and ovarian castration showed that natural aging method was better than that of ovarian castration method. Methods ： By observing the vaginal exfoliated cells, compare estrus cycle of the natural aging and ovarian castration mice. Observe the mice tail suspension test. Compare mice emotional state of natural aging and ovarian castration group. Detect and compare serum FSH, LH and E2 levels. Results ： The estrus cycle of natural aging and ovarian castration group disordered. The estrus frequency reduced. The tail suspension test of the each group improved significantly and the serum FSH and LH levels of two groups increased and E2 level decreased significantly. The hypothalamus KiSS - 1 and its receptor GPR54 mRNA expression increased significantly. Compared with ovarian castrated mice, the natural aging mice estrus was relatively longer, estrogen frequency decreased and the serum FSH, LH, KiSS - 1 and its receptor GPR54 mRNA levels in- creased and E2 level decreased significantly. Conclusion：Natural aging method and ovarian castration method both could establish the perimenopausal syndrome model in mice,nevertheless aging method was better than another.
