酸性pH环境对成骨细胞增殖和凋亡的影响 被引量:4

摘要 目的 :研究酸性pH环境对大鼠原代成骨细胞增殖和凋亡的影响。方法 :采用机械分离法及二次酶消化法获得新生SD大鼠成骨细胞。细胞经鉴定、培养、传代后,取第4代成骨细胞并随机分为3组,分别为培养基pH 6.4、6.8及7.4组。细胞在经过不同pH值的培养基处理12、24、36、48 h后,采用细胞计数法检测酸性环境对原代成骨细胞存活率的影响;MTT比色法检测原代成骨细胞在酸性条件下的增殖活性;Annexin V-PI染色法检测不同pH培养基对成骨细胞凋亡的影响。采用SPSS16.0软件包对数据进行统计学分析。结果:随着酸性培养基作用时间的延长和p H值的降低,实验组(p H 6.4、6.8)的细胞活性和细胞增殖均呈递减趋势,而细胞凋亡逐渐增加。各组细胞的细胞活性、增殖和凋亡在各个时间点均有显著差异(P<0.05)。结论:酸性环境能抑制大鼠成骨细胞增殖和分化,促进成骨细胞凋亡。 PURPOSE: To investigate the influence of acidic media with low p H value on the proliferation and apoptosis of rat osteoblasts. METHODS: Osteoblasts were separated from Sprague-Dawley neonatal rats by mechanical methods and second enzyme digestion. After primary culture, proliferation and identification of osteoblasts, the fourth generation of cells were exposed to different p H(6.4, 6.8, 7.4) with different time(12, 24, 36, 48 h). Cell viability was detected by cell count, cell proliferation was assessed by MTT assay and apoptosis was determined by flow cytometry. The difference between each group was analyzed with SPSS 16.0 software package. RESULTS: The viability and proliferation of rat osteoblasts in the experimental groups were suppressed gradually and the induction of apoptosis was increased greatly with the extension of time and decrease of p H value. There was significant difference on viability, proliferation and apoptosis among the three groups(P〈0.05). CONCLUSIONS: The viability and proliferation of osteoblasts were inhabited while the induction of apoptosis was promoted under acidic environment with low p H value.
