紫山药组培苗生根培养基优化与移栽驯化技术研究 被引量:1

摘要 紫山药组培苗生根诱导培养基优化组合不同诱导试管生根苗的炼苗(移栽)、炼苗基质、季节、容器影响成活的比较试验。试验结果表明:紫山药试管生根苗以1/2 MS+NAA mg/L+AC g/L的生根率高,达标试管移栽苗3~5 cm长、茎粗、根系发达、叶片大深绿;基质以珍珠岩:黄土=3:1;营养钵容器在当年的4-5月份温度15~25℃,湿度控制在70%~80%下的弱光照或搭阴棚下炼苗较佳。炼苗成活率达70%~85%。 The optimum compositions of root inducing medium,various matrices for acclimation and transplantation,the effects of seasons and types of matrix container on the survival rate of test-tube plantlets of purple yam were tested. The results showed:the high root inducing rate was obtained in the medium of 1/2 MS supplemented with NAA mg/L+AC g/L. The plantlets ,which reached to 3~5 cm height with strong stems,dark green leaves and well developed root systems,could be trans-planted ex vitro. During the period of April and May,the plantlets were transplanted into individual pot filled with pearlite:yellow earth (3:1) as matrix. The pots were then put in shady conditions with the temperature of 15~25℃ and the relative humidity of 70% ~80%,the acclimating survival rate could reach to 70% ~85%.
