小檗胺对人肝癌SMMC-7721细胞增殖作用及其机制的初步研究 被引量:4

摘要 目的:研究小檗胺对人肝癌SMMC-7721细胞增殖作用的影响及可能的机制。方法:经不同浓度小檗胺处理体外培养的肝癌SMMC-7721细胞,采用MTT法检测细胞增殖的变化;采用PI染色法检测肿瘤细胞周期分布的变化;采用RT-PCR和Western blot法检测肿瘤细胞中Cyclin B1、Cyclin D1、p21和p27的表达水平。结果:与溶剂对照组比较,不同浓度的小檗胺均可明显抑制人肝癌SMMC-7721细胞增殖。PI染色结果显示,小檗胺可明显地诱导肝癌细胞周期阻滞于G0/G1期(P<0.05)。RT-PCR和Western blot结果均显示,应用小檗胺处理肿瘤细胞后,p21和p27的表达明显升高,而Cyclin B1和Cyclin D1的表达水平明显下降(P<0.05)。结论:小檗胺能抑制人肝癌SMMC-7721细胞增殖,并使其细胞周期阻滞于G0/G1期,其机制可能与其上调p21和p27的表达和下调Cyclin B1和Cyclin D1的表达有关。 OBJECTIVE To investigate effects and possible mechanism of berbamine on proliferation of human liver cancer SMMC-7721 cells.METHODS After SMMC-7721 cells were incubated with various concentrations of berbamine,cell proliferation was assessed by MTT assay.Cell cycle distribution was detected by PI staining.Expression of Cyclin B1,Cyclin D1,p21 and p27 mRNA and protein was determined by RT-PCR and western-blot assay.RESULTS Compared with control group,berbamine at different concentrations could obviously inhibit proliferation of SMMC-7721 cells and induce cell cycle arrest at G0/G1phase(P〈0.05).RT-PCR and Western blot assay confirmed that berbamine could decrease expression of Cyclin B1 and Cyclin D1,and increase expression of p21 and p27(P〈0.05).CONCLUSION Berbamine can induce SMMC-7721 cell proliferation and cell cycle arrest at G0/G1 phase,which may be associated with down-regulation of Cyclin B1 and Cyclin D1,and upregulation of p21 and p27 levels.
