摘要
西方蜜蜂(Apis砌ZZ澹ra)的线粒体DNA(mitochondrial DNA,mtDNA)细胞色素氧化酶亚基-I(cytochrome C oxidase subunit I,COI)~细胞色素氧化酶亚基-II(COII)富含A+T非编码区(AT-rich noncoding region,NC)属于高变异DNA序列,既呈现mtDNA长度变异又呈现mtDNA序列变异,它作为mtDNA多态性标记广泛地应用于A.m.地理亚种的DraI酶限制性片段长度多态性(restriction fragment length polymorphism, DraI RFLP)分析和单核苷酸多态性( single nucleotide polymorphism, SNP)分析。在本文中,A.m.非洲世系mtDNA的28种DraI RFLP谱图被归纳为3种COI—COIINC单倍型类群,即P0、Q元件组成类群,P1、Q元件组成类群以及P2、Q元件组成类群;A.m.西部欧洲世系mtDNA的90种DraI RFLP谱图被归纳为1种COI—COIINC单倍型类群,即P、Q元件组成类群;A.砚东部欧洲世系mtDNA的25种SNP谱图被归纳为1种COI—COIINC单倍型类群,即Q元件组成类群;A.m.亚洲世系mtDNA的29种DraIRFLP谱图被归纳为3种COI—COIINC单倍型类群,即P0、Q元件组成类群,P0’、Q元件组成类群以及P1',Q元件组成类群。本文综述了这个研究进展。
Cytochrome C oxidase subunit I (COI)-COII ingtergenic sequence AT-rich noncoding region (COI-COII NC) is specific for mtDNA of Apis mellifera (A. m. ). This region has considerable DNA variability in both length and sequence, for which it has been used extensively as a polymorphic marker of mtDNA for analyzing DraI RFLP and SNP among A. m. geographic subspecies. Here, mtDNA of A. m. lineage in Africa, which are characterized by 28 DraI RFLP patterns, is induced to be composed of 3 COI-COII NC haplotype groups of A. m., i.e. the groups with both P and Q elements where P can also appear in two types of variations (P0 and P1); mtDNA of A. m. lineage in western Europe, which are characterized by 90 DraI RFLP patterns, is induced to be composed of 1 COI-COII NC haplotype group of A. m., i.e. the group with both P and Q elements; mtDNA of A. m. lineage in eastern Europe, which are characterized by 25 SNP patterns, is induced to be composed of 1 COI-COII NC haplotype group of A. m., i.e. the group only with Q element; mtDNA of A. m. lineage in western Asia, which are characterized by 29 DraI RFLP patterns, is induced to be composed of 3 COI-COII NC haplotype groups of A. m., i.e. the groups with both P and Q elements where P can also appear in two types of variations (P0'and P1').
