摘要
目的建立高效液相色谱-二极管阵列检测器(HPLC-DAD)法同时测定四季三黄丸(大黄、黄柏、黄芩等)中9种成分的含有量。方法分析采用Agilent SB-C18色谱柱(150 mm×4.6 mm,5μm);乙腈(A)-0.5%磷酸水溶液(B)为流动相,梯度洗脱;体积流量0.8 m L/min;柱温38℃;大黄酸、大黄素、大黄素酚和大黄素甲醚检测波长254 nm,栀子苷240 nm,黄芩苷、黄芩素和汉黄芩素278 nm,巴马汀345 nm;进样量10μL。结果栀子苷、黄芩苷、黄芩素、巴马汀、大黄酸、汉黄芩素、大黄素、大黄酚、大黄素甲醚分别在15~225μg/m L(r=0.999 8)、8~120μg/m L(r=0.999 3)、7~105μg/m L(r=0.999 6)、10~150μg/m L(r=0.999 3)、20~300μg/m L(r=0.999 5)、6~90μg/m L(r=0.999 7)、5~75μg/m L(r=0.999 6)、15~225μg/m L(r=0.999 5)、21~315μg/m L(r=0.999 4)范围内线性关系良好,平均回收率98.82%~100.85%,RSD均小于2.0%。结论该方法简单、准确、可靠,可用于四季三黄丸的质量控制。
AIM To establish a high-performance liquid chromatography with diode array detection (HPLC- DAD) method for the simultaneous determination of contents of nine constituents in Siji Sanhuang Pills ( a medica- tion for inflammatory condition of the skin and urinary system, containing Rhei Radix et Rhizoma, Phellodendri Chinensis Cortex, Scutellariae Radix, etc. ). METHODS The analysis was performed on a 38 ℃ thermostatic Agilent SB-C18 column (150 mm ×4. 6 mm, 5 μm), with the mobile phase comprising of acetonitrile (A) -0. 5% phosphate acid aqueous solution (B) flowing at 0. 8 mL/min for gradient elution. The detection wavelengths were set at 254 nm for rhein, emodin, chrysophanol and physcion, 240 nm for geniposide, 278 nm for baicalin, biaca- lein and wogonin, and 345 nm for palmatine, and the injection volume was 10 txL. RESULTS Geniposide, ba- icalin, biacalein, palmatine, rhein, wogonin, emodin, chrysophanol and physcion showed good linear relationships within the ranges of 15-225 μg/mL (r=0.999 8), 8-120 μg/mL (r=0.999 3), 7-105 μg/mL (r= 0.9996), 10-150 μg/mL (r=0.999 3), 20-300 μg/mL (r=0.999 5), 6-90 μg/mL (r=0.999 7) 5 -75 μg/mL (r =0. 999 6), 15 -225 μg/mL (r =0. 999 5) and 21 -315 μg/mL (r =0. 999 4), respectively. The average recoveries were 98.82% - 100. 85% with the RSDs of less than 2. 0% . CONCLUSION This sim- ple, accurate and reliable method can be applied to the quality control of Siji Sanhuang Pills.
