摘要
本研究在前期已获得的黄颡鱼Pelteobagrus fulvidraco转录组基础上,利用BLAST程序从中筛选获得黄颡鱼c Gn RH基因的候选序列,再通过氨基酸序列分析从中筛选获得c Gn RH的全长c DNA为261bp,编码86个氨基酸,其核心十肽的序列为QHWSHGWYPG,相对分子质量为9 755.5、理论PI值为9.02。序列同源比对发现:黄颡鱼与斑马鱼Danio rerio、草鱼Ctenopharyngodon idella、鲤Cyprinus carpio中c Gn RH氨基酸的相似性分别高达70.93%、68.60%和67.44%。用MEGA构建的黄颡鱼与其他27种硬骨鱼c Gn RH的Neighbour-joining进化树发现,黄颡鱼c Gn RH先与脂鲤科的墨西哥丽脂鲤Astyanax mexican共聚类,再依次与鲤科的斑马鱼、草鱼、黑头呆鱼Pimephales promelas、稀有鮈鲫Gobiocypris rarus等鱼的c Gn RH序列聚类。本研究首次筛选获得黄颡鱼的c Gn RH序列,并利用生物信息学软件进行鉴定分析,为后续的基因表达和c Gn RH调节生长生殖作用的研究提供序列资源及基础。
In previous studies, our group has obtained transcription data of yellow catfish Pelteobagrus fulvidraco, and in current study candidate sequences of c Gn RH gene were screened and sequenced using the blast program and amino acid sequence analysis. Results showed that full-length c DNA of c Gn RH was found to be 261 bp, encoding 86 amino acid sequences, with ten core peptides of QHW SHGWYPG, relative molecular mass of 9 755.5 and the theory PI of 9.02. The similarity of c Gn RH amino acids in yellow catfish was as high as 70.93% to zebrafish Danio rerio, 68.60% to grass carp Ctenopharyngodon idella, and 67.44% to common carp Cyprinus carpio. The neighbour-joining phylogenetic tree of c Gn RH amino acids in yellow catfish and 27 other teleosts was established using MEGA. The c Gn RH of yellow catfish was found to be co-clustered with Astyanax mexicanus in Characidae preferentially, then co-clustered with zebrafish, grass carp, Pimephales promelas and Gobiocypris rarus in Cyprinidae. The c Gn RH sequence of yellow catfish was screened first time, and analyzed and identified using bioinformatics. The findings provide foundation for gene expression subsequently, and necessary sequence resources for analysis of the regulation of c Gn RH in reproduction and growth.
