摘要
目的比较不同抗焦虑天然药物中桃叶珊瑚苷的含量,为抗焦虑天然药物的开发与应用提供实验依据。方法采用甲醇闪式提取方法对地黄、栀子、车前草、蜘蛛香及大王、密穗、头花、管花马先蒿等8种抗焦虑药物进行提取,利用高效液相色谱法分别对提取物中所含主要活性成分桃叶珊瑚苷进行定量研究。色谱条件为ZORBX-C18(4.6mm×150mm,5μm);流动相:甲醇-水溶液(5∶95);检测波长203nm;流速1ml/min;柱温25℃;进样量10μl。结果桃叶珊瑚苷线性范围为3.75~300.00μg/ml,加样回收率为98.67%,RSD为1.97%;地黄、栀子、车前草、蜘蛛香及大王、密穗、头花、管花马先蒿等8种天然药物中桃叶珊瑚苷的含量分布为0.016%、0.615%、0.060%、0.006%、0.097%、0.845%、0.677%、0.947%。结论管花、密穗、栀子、头花等药物中桃叶珊瑚苷含量较高,而地黄、蜘蛛香等药物桃叶珊瑚苷的含量较低,其成分含量有一定差异。闪式提取法可高效、快速提取环烯醚萜等成分。
Objective To compare the differences about the content of aucubin among natural medicines with anxiolytic effect and to provide experimental evidence for the development and application of these medicines. Methods The methanol extracts from eight natural medicines with anxiolytic effect were obtained by smashing tissue extraction technology and the contents of aucubin were determined by HPLC. The chromatographic column: ZORBX- C18( 4. 6mm × 150 mm,5μm),mobile phases: Me OH-H2O( 5∶ 95),detection wavelength: 203 nm,flow rate: 1. 0m L / min,column temperature: 25℃,volume of injection: 10μl.Results The method was linear within the range of 3. 75μg / m L ~ 300. 00μg / m L,the average recovery was 98. 67%,and the RSD was 1. 97%( n = 5). The contents of aucubin in eight medicinal plants were as follows: aucubins in Rehmannia glutinosa,Gardenia jasminoides,Plantago depressa,Valeriana jatamans,Pedicularis rex,Pedicularis densispica,Pedicularis cephalantha,Pedicularis siphonantha were 0. 016%,0. 615%,0. 060%,0. 006%,0. 097%,0. 845%,0. 677%,0. 947%. Conclusion There was a great difference in the contents of aucubin in eight natural medicines. The contents of aucubin in P. siphonantha,P. densispica,G. jasminoides,P. cephalantha were higher,while the contents of aucubin in R. glutinosa,V. jatamans lower. The iridoids can be extracted efficiently,rapidly by smashing tissue extraction method.
