胆管癌胆汁中糖蛋白的糖链结构特点 被引量:2

Characteristics of oligosaccharides of glycoproteins in bile from patients with bile duct carcinoma

摘要 目的 探讨与胆管癌相关的胆汁糖蛋白的糖链结构特点。方法 对 1 5例胆管癌胆汁及 1 0例良性胆道疾病的胆汁蛋白进行蛋白质电泳考马斯亮蓝及银染色 ,并用 4种识别不同糖链结构的辣根过氧化酶标记的凝集素 (ConA HRP、WGA HRP、LCA HRP、DSA HRP)进行凝集素印迹分析。结果 胆管癌胆汁和良性胆汁蛋白SDS -PAGE谱基本相同。ConA结合的胆管癌相关糖蛋白有 4种 (~ 1 38、~ 1 2 2、~ 1 0 8、~ 1 0 1kDa) ;WGA结合胆管癌相关糖蛋白 1种 (~ 2 0 1kDa) ;LCA结合胆管癌相关糖蛋白 1种 (~ 1 2 2kDa) ;DSA结合胆管癌相关糖蛋白 3种 (~ 2 0 1、~ 1 63、~ 1 2 2kDa)。结论 胆管癌胆汁中一些糖蛋白的糖链结构发生了改变 ,主要表现为唾液酸含量增加及天线数、高甘露糖型糖链增多 ,可能与胆管上皮的恶性转化有关。 Objective To study characteristics of oligosaccharides of bile duct carcinoma (BDC) related glycoproteins in bile. Methods Biliary proteins from 15 patients with BDC and 10 with benign biliary diseases (BBD) were separated by SDS PAGE with coomasie blue and silver staining. Lectin blotting was used for all samples by 4 lectins conjugated hoseradish peroxidase which can recognize different special oligosaccharides. Results No significant difference of protein separated by SDS PAGE was found between BDC and BBD groups. The major BDC related lectin binding glycoproteins in bile found by lectin blotting were: four of ConA binding glycoproteins of Mr ~138, ~122, ~108 and ~101 kDa; one of ~201 kDa WGA binding glycoprotein; one of ~122 kDa LCA binding glycoprotein; three of DSA binding glycoproteins of Mr ~201, ~163 and ~122 kDa. Conclusions Oligosaccharide structures of some glycoproteins in the BDC bile alter. The changes include addition of sialic acid, increase of antenna number and mannose rich type oligosaccharide. All these changes may be related to transformation of epithelial cells of bile duct.
