摘要
目的检测前列腺增生患者逼尿肌中P2X_1受体的表达,探讨嘌呤能神经受体P2X1受体对引起不稳定膀胱的作用。方法选择单纯膀胱流出道梗阻(BOO组)和逼尿肌不稳定(DI组)患者的膀胱组织标本各10例,采用免疫组化(SP法)检测其在逼尿肌及上皮中P2X_1受体的表达,并转为OD值进行半定量分析,采用Mann-Whitney U检验对结果进行分析。结果 P2X_1在膀胱逼尿肌和上皮中均可表达。与BOO组比较,P2X_1受体在DI组逼尿肌中的表达明显升高(P<0.05),而在上皮中的表达差异不显著。结论前列腺增生时,P2X_1可能介导嘌呤能神经信号,在神经源性和肌源性两方面导致逼尿肌不稳定。
Objective To detect the expression ofP2X1 in the detrusor muscle of benign prostatic hyperplasia(BPH)patients, and investigate the role of purine nerveP2X1 receptors in the occurrence of unstable bladder. Methods Ten patients suffered with benign prostatic hyperplasia with a urodynamically proved detrusor stablity(BOO group) and detrusor instablity(DI group) were selected respectively. The expression ofP2X1 receptors on the epithelium and in the detrusor muscle of BPH patients were detected by immunohistochemical method(SP method) and were transferred to the OD value for the semi-quantitative analysis. The results were tested by the Mann-Whitney U test. ResultsP2X1 receptor is expressed both on the epithelium and in the detrusor muscle of BPH patients. The expression ofP2X1 receptor in detrusor muscle of BPH patients in DI group are significantly higher than that in BOO group(P〈0.05). However, the expression ofP2X1 receptor on the epithelium of BPH patients have no significant difference in the two groups(P〈0.05). ConclusionP2X1 may mediate signals of purinergic nerves, and cause unstable detrusor muscle from aspects of myogenic and neurogenic sides.
