黄芪甲苷对局灶性脑缺血再灌注损伤大鼠氧化应激的影响 被引量:12

The Effects of Astragalosides on Oxidative Stress in Rats with Focal Cerebral Ischemia-reperfusion Injury

摘要 目的观察黄芪甲苷对局灶性脑缺血再灌注大鼠氧化应激损伤的影响并探讨其可能的作用机制。方法采用线栓法制备局灶性脑缺血再灌注损伤大鼠模型,随机分为模型组、黄芪甲苷小(10 mg/kg)、中(20 mg/kg)、大剂量组(40 mg/kg)和银杏叶提取物组(200 mg/kg),各20只,另取20只同龄大鼠作为假手术组;各治疗组于再灌注30 min前腹腔注射给药。再灌注24 h后,行神经功能评分,测定脑组织含水量和脑梗死体积;检测脑组织中抗氧化酶[过氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、过氧化氢酶(CAT)]活性和丙二醛(MDA)含量,测定血清中肌酸激酶(CK)、乳酸脱氢酶(LDH)含量,苏木精-尹红(HE)染色法观察脑组织形态学变化;末端标记(TUNEL)法观察神经细胞凋亡状况并计算凋亡指数(AI),Western blotting法测定NF-κB蛋白表达。结果与模型组比较,黄芪甲苷中、大剂量组大鼠神经功能评分显著降低,含水量和脑梗死体积显著降低;脑组织中SOD、GSH-Px、CAT活性显著升高且MDA含量显著降低,血清中CK、LDH含量显著降低;黄芪甲苷各治疗组大鼠脑组织病变和神经细胞凋亡状况明显好转,均以大剂量组效果最为显著,黄芪甲苷中、大剂量组AI显著降低,脑组织中NF-κB蛋白表达显著下调,差异均具有统计学意义(P<0.05或P<0.01)。结论黄芪甲苷具有抑制局灶性脑缺血再灌注损伤大鼠氧化应激损伤的作用,作用机制可能与黄芪甲苷能够有效改善抗氧化酶活性、下调NF-κB蛋白表达、抑制神经细胞凋亡有关。 Objective: To investigate the effects of Astragalosides on oxidative stress in rats with focal cerebral ischemia-reperfusion injury. Methods: One hundred rat models with focal cerebral ischemic reperfusion made by suture method were randomly divided into five groups: the model control group,Astragalosides(10,20,40 mg/kg)treated groups and Ginkgo biloba extract(200 mg/kg) treated group,and another 20 same-aged rats were selected as the sham group. 30 min before operation,the drugs were given by intraperitoneal injection. Twenty-four hours after reperfusion,the neurological deficits,ratio of infarct volume,water content of the brain were evaluated;the activity of SOD,GSH-Px,CAT and the content of MDA in brain tissue were determined;the content of CK,LDH in serum were determined;the histopathological changes and neurocyte apoptosis were observed,and the AI were determined;the expression of NF-κB protein was detected by Western blot and Semi-quantitative analysized. Results: Compared with the model control group,the neurological scores of Astragalosides(20,40 mg/kg) treated groups were significantly decreased,and the ratio of infarct volume and brain water content were significantly decreased;the activity of SOD,GSH-Px,CAT in brain tissue were significantly increased and the content of MDA were significantly decreased,and the content of CK,LDH in serum were significantly decreased;the histopathological changes and neurocyte apoptosis of Astragalosides treated groups were significantly improved,and the AI of Astragalosides(20,40 mg/kg) treated groups were significantly decreased;the expression of NF-κB protein was significantly down-regulated;all of the difference above were significant(P 0.05 or P 0.01). Conclusion: Astragalosides has inhibitive effects on the oxidative stress in rats with focal cerebral ischemia- reperfusion injury,which is perhaps related to its pharmacological effects of enhancing the activity of antioxidant enzymes,regulating the expression of NF-κB protein and depressing neurocyte apoptosis.
