摘要
目的研究膀胱癌5637细胞中CDH13表达下调后对细胞迁移、侵袭和PI3K/Akt表达的影响以及它们间的关系。方法应用RNA干扰技术抑制人膀胱癌5637细胞株中CDH13的表达,划痕实验检测细胞迁移能力,Transwell小室法检测细胞侵袭能力,Western blot检测CDH13、PI3K及Akt的表达水平。结果膀胱癌5637细胞中CDH13表达下调后细胞迁移和侵袭能力增强,PI3K及Akt的表达增加。结论膀胱癌细胞中CDH13表达下调可能通过激活PI3K/Akt通路促进肿瘤细胞的迁移和侵袭。
Objective The aim of this study was to investigate the influence of down-regula-tion expression of CDH13 on the migration and invasion of bladder cancer cell and PI3K/Akt expres-sion,as well as the relationship between them. Methods RNA interference was used to inhibit CDH13 expression in 5637 cells,wound healing was used to examine cell migration,Transwell was used to evaluate cell invasion,the expression of CDH13,PI3K and Akt was examined by Western blot. Results The bladder cancer cell migration,invasion and PI3K/Akt expression was increased after the down-regulation of CDH13. Conclusions Down-regulation expression of CDH13 in blad-der cancer cell maybe promotes tumor cell migration and invasion through PI3K/Akt pathway.
