摘要
目的探讨复荣通脉胶囊对糖尿病大鼠坐骨神经Bax及Bcl-2表达的影响。方法将60只大鼠分为正常组(10只)和造模组(50只)。造模组大鼠制备糖尿病模型后随机抽取40只,将其分为糖尿病组(10只)、复荣通脉低剂量组(10只)、复荣通脉中剂量组(10只)和复荣通脉高剂量组(10只),分别给予0.7、1.4、2.8 g/kg复荣通脉灌胃,正常组和糖尿病组给予等量纯净水灌胃。灌胃8周后,留取大鼠坐骨神经制备病理切片,SP法免疫组化染色,观察各组Bax及Bcl-2表达情况。结果与正常组比较,糖尿病组及复荣通脉低、中、高剂量组Bax表达明显升高,Bcl-2表达明显下降(P<0.05);复荣通脉低剂量组Bax及Bcl-2表达与糖尿病组比较,差异均无统计学意义(P>0.05);复荣通脉中、高剂量组Bax表达明显降低,Bcl-2表达明显升高(P<0.05);与复荣通脉低剂量组比较,复荣通脉中、高剂量组Bax表达明显降低,Bcl-2表达明显升高(P<0.05);复荣通脉高剂量组Bax及Bcl表达与复荣通脉中剂量组比较,差异均无统计学意义(P>0.05)。结论复荣通脉胶囊可抑制糖尿病大鼠坐骨神经Bax的表达和Bcl-2表达下降,具有抑制糖尿病大鼠坐骨神经细胞凋亡的作用。
Objective To explore the the influence of FuRongTongMai Capsules on sciatic nerve expression of Bax and Bcl-2 in rats with diabetes mellitus (DM). Methods 60 rats were randomly divided into two groups: normal control group (NC, n = 10), build modle group (n = 50). After the diabetes mellitus (DM) rats were made, 40 of diabetes rats were randomly divided into DM group (n = 10), FuRongTongMai low dosage group (FRL) (n = 10), FuRongTongMai mid- dle dosage group (FRM) (n = 10), FuRongTongMai high dosage group (FRH) (n = 10), and they were given the 1.4, 0.7, 2.8 g/kg FuRongTongMai Capsules by gavage respectively, and rats of the control group and DM group were given the same dose of purified water daily. 8 weeks after lavage, sciatic nerve was took down for pathology slice, the expression of Bax and Be1-2 was observed after SP immunohistochemistry staining. Results Compared with NC group, sciatic nerve expression of Bax in the DM group, FRL group, FRM group, and FRH group was significantly increased and Bcl-2 was significantly decreased (P 〈 0.05); compared with DM group, expression of Bax and Bcl-2 in the FRL group had no significantly (P 〉 0.05), expression of Bax in the FRM group and FRH group was significantly decreased and Bcl-2 was significantly increased (P 〈 0.05); compared with FRL group, expression of Bax in the FRM group and FRH group was significantly decreased and Bcl-2 was significantly increased (P 〈 0.05); compared with the FRM group, expression of Box and Bcl-2 in the FRH group had no significantly (P 〉 0.05). Conclusion FuRongTongMai Capsules can inhibit the expression of Bax and the decreasing of Bcl-2 in sciatic nerve of diabetes rats, have the function of inhibit sciatic nerve apoptosis in diabetes rats.
