摘要
目的:探讨葛根素对雷帕霉素造成人脐静脉内皮细胞(HUVEC)损伤的影响及其可能机制。方法:培养HUVEC并分组:对照组,葛根素组,雷帕霉素组,葛根素干预组。各组培养48h后,检测各组HUVEC的增殖,NO浓度及凋亡率的情况。结果:与对照组比较,葛根素组NO浓度明显升高(P<0.05),HUVEC凋亡率明显降低(P<0.01);葛根素干预组NO浓度明显降低(P<0.01);雷帕霉素组可抑制HUVEC增殖,NO浓度明显降低,HUVEC凋亡率明显升高(P<0.01)。与雷帕霉素比较,葛根素组NO浓度明显升高,HUVEC凋亡率明显降低(P<0.01);葛根素干预组促进HUVEC增殖,NO浓度明显升高,HUVEC凋亡率明显降低(P<0.05,P<0.01)。结论:葛根素对雷帕霉素造成HUVEC损伤具有一定的拮抗作用。
Objective:To study the effect of puerarin on injury of human vascular endothelial cells(HUVEC)induced by rapamycin in vitro and to explore its regulation mechanisms.Method:The HUVEC were cultured and divided into four groups:normal group,puerarin group,rapamycin group and puerarin pretreat group.After 48 h culture,the proliferation of HUVEC,the level of NO,and the rate of apoptosis were detected.Result:Compared with normal group,NO levels were significantly increased in puerarin group(P〈0.05),meanwhile,the rate of apoptosis was deceased(P〈0.01);however,NO levels were markedly reduced in puerarin pretreat group(P〈0.01).Rapamycin inhibited the proliferation of HUVEC,reduced NO levels but increased the rate of apoptosis(P〈0.01).Compared with rapamycin group,the cells treated with puerarin had higher level of NO and proliferation but lower rate of apoptosis(P〈0.01).Conclusion:Puerarin can attenuate HUVEC injury induced by rapamycin.
